人参皂苷Rg3抑制人骨肉瘤细胞增殖及其分子机制研究

研究背景:骨肉瘤是发源于间充质干细胞的恶性肿瘤。骨肉瘤的发病年龄呈现双峰分布,常发病于青少年或老年人。由于大多数的骨肉瘤患者在诊断时时就有转移或微转移性,故几乎所有患者在手术切除的之前或之后都会接受多种药物联合化疗。患者往往耐受度较差,且复发几率较高。人参是一种原产于东北亚的草本植物,在东亚地区的传统医学中有着广泛的应用。作为人参中主要的活性物质之一,人参皂苷Rg3的抗肿瘤作用在近年来越来越引起人们的重视。人参皂苷Rg3的甾体结构使得它可以与细胞膜、膜结合离子通道、以及胞内和胞外的受体相互作用来改变细胞的转录水平以来发挥抗肿瘤作用。研究方法:本实验应用MTT实验、流式细胞术、细胞划痕实验、Transwell细胞侵袭实验检测了人参皂苷Rg3对骨肉瘤细胞系MG-63、HOS和U2OS增殖、凋亡、迁移、侵袭的影响和对细胞周期作用。我们又进一步选择MG-63细胞作为研究对象,应用western blot、定量PCR、流式细胞术、小干扰RNA等实验手段进一步探究人参皂苷Rg3诱导细胞自噬、凋亡以及细胞周期阻滞的具体作用机制。实验结果:MTT实验发现人参皂苷Rg3以剂量依赖的方式抑制三种骨肉瘤细胞...
