熊去氧胆酸对胆汁酸肠肝循环动力学的作用研究

背景及目的近年来,科学家发现胆汁酸(bile acids,BAs)不仅可以作为乳化剂促进脂类营养物质、脂溶性维生素的吸收,还可以作为信号分子通过调节多种受体参与糖、脂、能量代谢并维持自身平衡。胆汁酸的核受体之一法尼酯X受体(farnesoid x receptor,FXR)在胆汁酸的肠肝循环以及维持胆汁酸稳态方面有重要调节作用,然而不同胆汁酸对FXR的作用以及对胆汁酸肠肝循环动力学的影响目前尚未有明确的结论。熊去氧胆酸(ursodeoxycholic acid,UDCA)作为胆汁酸的一种,具有利胆、抗炎、细胞保护的特性,是被美国食品药品监督管理局(food and drug administration,FDA)认可的治疗原发性胆汁性胆管炎(primary biliary cholangitis,PBC)的药物。过去的研究认为UDCA可以通过FXR介导正向调控肝脏毛细胆管侧胆汁酸转运体的表达;然而有研究表明UDCA在人体内起到抑制FXR的作用,这使得UDCA促进肝胆汁酸分泌的作用机制仍有待明确。除了应用于胆汁阻塞性疾病,UDCA及其衍生物在其他肝脏类疾病中也有所应用,针对近年来发病率不...
