苏尼特羊BMPR-IB、BMP15和GDF9基因与多胎性状关联性分析及BMP15基因突变检测
[Abstract]:Sunita sheep is a Mongolian sheep breed, which has the advantages of cold tolerance, easy grazing and delicious meat quality, but its production performance is low. In this study, 300 Sunita sheep (including 188 single lambs and 112 double lambs) were selected as control, and 50 small tail Han sheep and 50 Hu sheep were selected as control, the total number of the whole research population was 400. Polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) and DNA sequencing technique were used to analyze the effects of different mutation sites of BMPR-IBBMP15 and GDF9 genes on the fertility traits of Sunita sheep. The aim is to provide theoretical support for improving the fecundity of Sunita sheep. The main results are as follows: (1) FecB mutation (A746G) was found in some Sunita sheep with two lambs, the mutation types were heterozygous (AG) and homozygous (GG), the genotype frequencies were 0.089 and 0.009A and G allele frequencies were 0.946 and 0.054, respectively; The population of Niett sheep showed low polymorphism at PIC-0.097 at this locus. There was no polymorphism at this locus in single lamb Sunita sheep population. The frequency of G allele in single lamb Sunita sheep was significantly lower than that in double lamb Sunita sheep (P0.01), and the correlation analysis showed that the frequency of G allele in Sunita sheep was related to the rate of double lambs (P0. 001). (2). No mutations were found at the five FecXHG and FecXI loci of the BMP15 gene. All were homozygous wild type. (3) there was no polymorphism at the four loci of FecGH FecGFGE and G4 on the GDF9 gene in the selected Sunita sheep population. All of them were homozygous wild type. (4) the following mutations were found in the intron of part of Sunita sheep BMP15 gene by sequencing. The following mutations occurred in the intron of the BMP15 gene: 10 A115G, G117A, C342A, G419A, A1168G and G2129T. The results show that the FecB mutation site can be used as a molecular marker to improve the lambing rate of Sunita sheep. However, the molecular genetic mechanism of double lamb traits of Sunita sheep needs to be further studied.
