【摘要】:Ⅰ型超敏反应又称速发型超敏反应,它的主要特征:发病快,机体出现生理功能性紊乱,有明显个体差异和遗传背景。常见病有:青霉素过敏、花粉病、支气管哮喘、过敏性鼻炎、特应性皮炎、食物过敏症等。以哮喘为例,它是世界上最常见的慢性疾病之一。目前全世界有近2亿患者,我国患病率接近1%,患病人数在1千万以。Ⅰ型超敏反应主要由人免疫球蛋白E(human immunoglobulin E,IgE)介导发生,IgE是正常人血清中含量最少的Ig,血清浓度极低,约为5×10~(-5)mg/ml,主要由黏膜下淋巴结组织中的浆细胞分泌。IgE为亲细胞抗体,其CH2和CH3结构域可与肥大细胞、嗜碱性粒细胞上FC ε RI高亲和力的结合,引起Ⅰ型超敏反应。现今抗IgE疗法即用抗IgEmAb阻断IgE或阻断它的合成为包括哮喘在内的过敏性疾病患者提供了极为有效的治疗手段。但是,这些药物存在有用药量大、需长期反复使用、易引起超敏反应的缺陷。表位肽疫苗就是指用短的、合成的表位肽进行免疫治疗,只需少量注射就可以诱发产生抗体,可克服抗体及可溶性受体药物的缺陷。因此IgE表位肽疫苗是治疗Ⅰ型超敏反应性疾病的一条有效途径。噬菌体呈现肽库是近年来发展很快的一项技术,它已成为为已知分子寻找相应配体的有效工具。因此本试验希望利用噬菌体筛选技术筛选得到IgE表位
[Abstract]:Type I hypersensitivity is also known as the rapid hypersensitivity, its main characteristics: rapid onset, physical functional disorder, obvious individual difference and genetic background. Common diseases are: penicillin allergy, hay fever, bronchial asthma, allergic rhinitis, atopic dermatitis, food allergy, and so on. As an example of asthma, it is one of the most common chronic diseases in the world. At present, there are nearly 200 million patients all over the world. The prevalence rate of China is close to 1%, and the number of diseases is 10 million. Type I hypersensitivity is mainly mediated by Human Immunoglobulin E (human immunoglobulin E, IgE). IgE is the least Ig in normal human serum, and its serum concentration is very low, which is about 5 10~ 10~ (-5) mg/ml, which is mainly secreted by plasma cells in submucosal lymph node tissue. IgE is a pro cell antibody, and its CH2 and CH3 domains can be combined with the high affinity of FC epsilon RI on mast cells and basophils, causing type I hypersensitivity. Anti - IgE therapy today is an extremely effective treatment for patients with allergic diseases, including asthma, using anti IgEmAb blocking IgE or blocking its synthesis. However, these drugs have a large amount of useful drugs, long-term repeated use, easy to cause hypersensitivity defects. Epitope peptide vaccine refers to immunotherapy with short and synthetic epitope peptides. Only a small amount of injection can induce antibodies, which can overcome the defects of antibodies and soluble receptor drugs. Therefore, the IgE epitope peptide vaccine is an effective way for the treatment of type I hypersensitivity disease. Phage display peptide library is a fast developing technique in recent years. It has become an effective tool for finding the corresponding ligands for known molecules. Therefore, we hope to use phage screening technique to screen the IgE epitopes
