雌二醇和孕酮对小鼠心肌细胞超微结构的影响

【摘要】:目的 通过观察苯甲酸雌二醇、乙酸羟孕酮以及二者联用替代疗法短期和长期作用对卵巢切除小鼠心肌细胞的数量、超微结构形态学改变,从而探讨苯甲酸雌二醇、乙酸羟孕酮对机体心血管系统的作用机制。 方法 采用健康雌性昆明小鼠60只,随机分为五组:一组为保留卵巢空白对照组(INT),未加任何处理;其余四组去势后,分别注射生理剂量的苯甲酸雌二醇(OVX+EB组)、乙酸羟孕酮(OVX+HPC组)、苯甲酸雌二醇、乙酸羟孕酮联用(OVX+EB.HPC组)以及麻油(麻油对照组,OVX组)。每组12只,药物注射从小鼠40天开始,每5天注射药物一次,30次为一个疗程,共150天。分别与半个疗程和疗程结束时各组取材制备电镜标本,代表短期治疗组和长期治疗组,电镜下观察各组心肌细胞超微结构的形态学改变。 结果 不同治疗组间比较:形态学观察:空白对照组(INT)电镜下心肌细胞超微结构未见改变,符合正常心肌的组织形态。雌二醇组(OVX+EB)、孕酮组(OVX+HPC)、雌二醇加孕酮组(0VX+EB.HPC)和去卵巢组(OVX)心肌细胞超微结构有不同程度的改变。去卵巢组与对照组比较改变最为明显,以凝集性改变为主。雌二醇组、孕酮组、雌二醇加孕酮组超微结构改变程度介于对照组与去卵巢组之间。取闰盘形态变化作定性指标进行统计学处理,短期治疗组与长期治疗组统计结果显示:OVX组与其它四组比较差别有统计学意义(P0.01);OVX+EB组、OVX+HPC组和OVX+EB.HPC组与INT组比较差别没有统计学意义(P0.05);OVX+EB组、OVX+HPC组和OVX+EB.HPC组与OVX组比较差别有统计学意义(P0.01);OVX+EB组、OVX+HPC组和
[Abstract]:Objective to investigate the morphologic changes of cardiomyocytes in ovariectomized mice by observing the effects of estradiol benzoate, hydroxyprogesterone acetate and the combination of them on the number and ultrastructure of cardiomyocytes in ovariectomized mice. Mechanism of Action of Hydroxyprogesterone Acetate on Cardiovascular system. Methods Sixty healthy female Kunming mice were randomly divided into five groups: one group was treated without any treatment, the other four groups were ovariectomized. 12 rats in each group were injected with estradiol benzoate OVX EB group, OVX HPC group with hydroxyprogesterone acetate, estradiol benzoate and hydroxyprogesterone acetate combined with OVX EB.HPC group, and sesame oil with 12 rats in each group. Every 5 days, 30 times of drug injection were used as a course of treatment for 150 days. Electron microscopic specimens were taken from each group at the end of half course and treatment course respectively, representing the short-term treatment group and the long-term treatment group. The ultrastructural changes of myocardial cells in each group were observed under electron microscope. Results comparison between different treatment groups: morphological observation: the ultrastructure of cardiomyocytes in the blank control group was not changed under electron microscope. The ultrastructure of cardiomyocytes in oestradiol group, progesterone group, estradiol plus progesterone group and ovariectomized group were different. The ultrastructural changes of estradiol group, progesterone group and estradiol plus progesterone group were between control group and ovariectomized group. The statistical results of short-term treatment group and long-term treatment group showed that there were significant differences between the two groups in comparison with the other four groups. There was no significant difference between the OVX EB.HPC group and the INT group and the OVX HPC group, the OVX EB.HPC group and the OVX group, and the OVX HPC group and the OVX EB.HPC group were not significantly different from the other four groups. The difference between the two groups was statistically significant (P 0.01). OVX HPC group and OVX HPC group were significantly different from those in EB group.
