【摘要】:免疫球蛋白(IgG)是免疫系统中的重要分子,以往经典的免疫学理论认为IgG都是B淋巴细胞和浆细胞分泌的,而新近的研究表明上皮性肿瘤也能分泌IgG。同时以往的研究已证实,在肿瘤细胞中,热休克蛋白(HSP)高表达。我们在实际工作中发现IgG和HSP70的免疫组化染色表现非常相似,于是我们设想IgG和HSP70也许有相互关系,并在肿瘤免疫中互相作用。 本课题旨在探讨:肿瘤细胞分泌的IgG与热休克蛋白70(HSP70)的共定位关系,并初步提出在肿瘤细胞内可能存在细胞内免疫机制,,讨论IgG和HSP70在细胞内免疫系统中的作用。目的:1.制备IgG的多克隆抗体;2.制备IgG的单克隆抗体;3.研究证实IgG和HSP70在肿瘤细胞中的共定位关系,为肿瘤细胞内免疫机制的存在提供初步证据。方法:1.用纯化的IgG以常规多克隆抗体制备方法制备兔抗人IgG的多克隆抗体,并纯化并对抗体特性进行鉴定;2.以纯化的IgG常规免疫、常
[Abstract]:Immunoglobulin G (IgG) is an important molecule in the immune system. The classical immunological theory holds that IgG is secreted by B lymphocytes and plasma cells, but recent studies have shown that epithelial tumors can also secrete IgG.At the same time, previous studies have confirmed that IgG can be secreted by B lymphocytes and plasma cells. In tumor cells, heat shock protein (HSP) is highly expressed. We found that IgG and HSP70 showed very similar immunohistochemical staining, so we assumed that IgG and HSP70 might be related to each other and interact with each other in tumor immunity. The aim of this study was to investigate the co-localization of IgG secreted by tumor cells and heat shock protein 70 (HSP70), and to suggest that there may be intracellular immune mechanisms in tumor cells. To discuss the role of IgG and HSP70 in the cellular immune system. Objective 1. To prepare the polyclonal antibody to IgG. 2. To prepare the monoclonal antibody against IgG. The co-localization of IgG and HSP70 in tumor cells was confirmed.
