【摘要】: 目的:明确一种人乳头瘤病毒(human papillomavirus,HPV)外壳蛋白L1的B细胞共同表位短肽作为HPV疫苗发展的可能性。 方法:通过对HPV-16、18、6和11型等几种主要致病型别HPV外壳蛋白L1的三维结构及其氨基酸序列的比较和分析,构建HPV外壳蛋白L1的B细胞共同表位短肽疫苗,并以该短肽对动物进行免疫接种诱导抗短肽抗血清,再用酶联免疫吸附测定(enzyme-linked immunosorbent assay, ELISA)、十二烷基磺酸钠-聚丙酰胺凝胶电泳(SDS-PAGE)、免疫印迹(Western blot)及免疫组化(immunohistochemistry)等方法对短肽和抗短肽抗血清的免疫学特性进行检测。 结果:短肽与Furend`s完全佐剂合用免疫动物可诱导高滴度的抗短肽抗血清,并且该抗血清能参与多型别HPV交叉中和反应。 结论:HPV外壳蛋白L1的B细胞共同表位短肽可以作为HPV疫苗候选物。
[Abstract]:Aim: to identify the possibility of the development of a human papillomavirus (HPV) coat protein L1 B cell epitope short peptide as a HPV vaccine. Methods: by comparing and analyzing the three dimensional structure and amino acid sequence of HPV coat protein L1, the common epitope short peptide vaccine of HPV coat protein L1 was constructed. The anti-short peptide antiserum was induced by immunizing animals with the short peptide. The immunological properties of short peptide and anti-short peptide antiserum were detected by enzyme-linked immunosorbent assay, ELISAA, sodium 12 alkyl sulfonate-polyacrylamide gel electrophoresis (SDS-PAGEG) and immunohistochemical histochemistry. etc. Results: the combination of short peptide and Furend's complete adjuvant could induce high titer of anti short peptide antiserum, and the antiserum could participate in cross neutralization reaction of multi type HPV. Conclusion the B cell common epitope short peptide of 1: HPV coat protein L 1 can be used as a candidate for HPV vaccine.
