骨髓多能成体祖细胞向表皮细胞分化的研究

【摘要】: 本研究通过克隆化培养,成功的从成年大鼠骨髓中分选出较为均一的具有多系分化潜能的多能成体祖细胞(MAPCs)群体。首次系统比较了骨髓中间充质干细胞(MSCs)和MAPCs在生物特性及分化潜能上的异同点。结果证实,MAPCs可能是MSCs更早期的一类远祖细胞;体外模拟MAPCs向皮肤上皮细胞分化的微环境,首次建立了MAPCs向表皮细胞体外分化模型,实现MAPCs向表皮样细胞的转型;在体观察了皮肤局部损伤微环境对MAPCs的迁移和诱导分化作用。结果提示MAPCs可定向迁移受损皮肤表面及皮肤附属器毛囊周围,并通过转化为表皮细胞参与受损皮肤的愈合。该工作不仅丰富了MAPCs可塑性基础理论研究,而且为指导细胞移植治疗应用到临床实践提供了一定的理论依据。
[Abstract]:In this study, a more homogeneous multifunctional adult progenitor cell (MAPCs) with multilineage differentiation potential was successfully isolated from adult rat bone marrow by cloning culture.Bone marrow mesenchymal stem cells (MSCs) and MAPCs were compared in biological characteristics and differentiation potential for the first time.The results showed that MPCs might be an earlier type of MSCs progenitor cells, and mimic the microenvironment of MAPCs differentiation into skin epithelial cells in vitro. The model of MAPCs differentiation into epidermal cells was established for the first time to realize the transition of MAPCs to epidermoid cells.The effects of local skin damage on MAPCs migration and differentiation were observed in vivo.The results suggest that MAPCs can migrate the damaged skin surface and the skin adnexa around the hair follicle and participate in the healing of the damaged skin by transforming into epidermal cells.This work not only enriches the basic theory of MAPCs plasticity, but also provides a theoretical basis for the application of cell transplantation therapy to clinical practice.
