甲状腺激素对大鼠睾丸间质细胞再生的影响

【摘要】: 目的:应用硝基蓝四唑盐(NBT)法、HE染色和磁分离均相酶联免疫法等方法观察睾丸间质细胞(Leydig cells, LCs)被二甲磺基乙烷(ethane dimethane sulphonate,EDS)破坏后在甲状腺机能亢进和甲状腺机能减退情况下的再生情况,探讨甲状腺激素对成熟大鼠LCs再生的影响。 方法:选用90日龄雄性SD大鼠54只,EDS一次性腹腔注射(85mg/kg)破坏LCs。将54只EDS处理后的大鼠随机分为三组:甲状腺机能亢进组、甲状腺机能减退组和对照组,每组18只。 甲状腺机能亢进组大鼠在注射EDS当日起每天皮下注射T3(60μg/kg)一次,造成甲状腺机能亢进模型;甲状腺机能减退组大鼠在注射EDS当日起每天用50mg丙硫氧嘧啶水溶液灌胃一次,建立甲状腺机能减退模型;对照组大鼠在注射EDS当日起每天皮下注射含0.025 M NaOH的生理盐水0.8 ml。 分别在注射EDS后的第7天、14天和21天麻醉大鼠、取材,进行如下指标的观察:①LCs的再生情况:取睾丸-70℃速冻,Leica冰冻切片机切片,片厚15μm,NBT法显示LCs内的3β-HSD酶,判定LCs的再生情况。②LCs的功能:取睾丸组织体外培养,加入黄体生成素(LH)后,采用磁酶免法检测上清液中睾酮的含量;下腔静脉取血,离心取血清,磁酶免法检测血清中睾酮的含量。③LCs的形态和数
[Abstract]:Objective: to observe the hyperthyroidism and hypothyroidism of Leydig cells (LCs) in testicular interstitial cells after the destruction of Leydig cells (LCs) by dimethylsulfoethane dimethane sulphonate EDSs by means of HE staining and magnetic separation homogenous enzyme linked immunosorbent assay (Elisa) with nitro blue tetrazolium (NBT). Methods: 54 male SD rats aged 90 days were injected intraperitoneally with single dose of 85 mg / kg to destroy LCs.
