糠秕马拉色菌胞外蛋白酶活性测定

【摘要】:目的:①检测糠秕马拉色菌胞外蛋白酶活性;②研究菌丝相及酵母相蛋白酶活性的差异;③蛋白酶活性的影响因素。 方法:选用糠秕马拉色菌标准株,接种于菜籽油液体培养基和菌丝液体培养基中振荡培养,,诱导糠秕马拉色菌菌丝相及酵母相生成,收集培养上清液。用全脂牛奶平板法、Folin-酚法、QuantiCleave蛋白酶试剂盒测定菌丝相及酵母相糠秕马拉色菌胞外蛋白酶的活力,并检测温度、pH值对蛋白酶活性的影响。 结果:①全脂牛奶平板法:培养第7天菌丝相上清液蛋白酶活性(12.5±1.2mm)高于酵母相(9.25±0.71mm)(p<0.05);菌丝相上清液蛋白酶活性随培养天数延长而增大;25℃和37℃两种温度对上清液蛋白酶活性无明显影响(P>0.05);②Folin-酚法:上清液蛋白酶活性随培养天数、菌落计数、菌丝诱导率增加而增大,培养液pH值呈下降趋势;同一天的菌丝相上清液蛋白酶活性明显高于酵母相(p<0.05);三种温度(25℃、37℃、49℃)及三种pH值(3.8、5.6、7.4)对上清液蛋白酶活性无明显影响;温度37℃、pH5.6时菌丝相上清液蛋白酶活性最高;③QuantiCleave蛋白酶试剂盒:菌丝相及酵母相上清液中均含有蛋白酶且
[Abstract]:Objective to detect the extracellular protease activity of Malassezia furfur by 1: 1 and to study the factors influencing the protease activity in mycelia and yeast. Methods: the standard strain of Malassezia furfuris was inoculated in the liquid medium of rapeseed oil and the liquid medium of hyphae to induce the formation of hyphae and yeast phase of Malassezia furfurum and the supernatant of culture was collected. The extracellular protease activity of Malassezia furfurum in hyphal phase and yeast phase was determined by Folin- phenol QuantiCleave protease kit, and the effect of pH value on the activity of protease was detected. Results: the protease activity of supernatant of mycelium phase on the 7th day was higher than that of yeast phase (9.25 卤0.71mm)(p < 0.05mm), and the protease activity of supernatant of mycelia phase increased at 25 鈩
