脑膜炎大肠杆菌K1株新基因cglE的克

【摘要】:细菌性脑膜炎是中枢神经系统(central nerve system,CNS)最常见最严重的感染性疾病。尽管抗生素的广泛使用,但细菌性脑膜炎的发病率仍有5%-40%,而幸存者神经系统后遗症的发生率高达30%。细菌性脑膜炎的高感染率和死亡率的主要原因之一是由于其发病机制还不清楚。 大肠杆菌(Escherichia coli E.coli)是导致新生儿脑膜炎最常见的革兰氏阴性杆菌。已知大部分发生在新生儿中的细菌性脑膜炎是细菌血源性传播的结果。进入中枢神经系统是细菌性脑膜炎发生的必要条件,但关于血循环中的大肠杆菌如何穿透由脑微血管内皮细胞(brain microvascular endothelial cells,BMECs)等组成的血脑屏障(blood-brain barrier,BBB),其机制仍待研究。目前在脑膜炎大肠杆菌感染组学的研究中,已经确定部分基因与细菌穿透血脑屏障有关。通过TnphoA突变实验发现基因ibeA在细菌侵袭BMEC过程中发挥着重要作用,且为毒力株E.coli K1株所独有的。包含着ibeA的基因岛命名为GimA(a Genetic island of meningitic E.coli containing ibeA)。GimA全长20.3kb,其GC含量(46.2%)与E.coli基因组的GC含量(50.8%)不同,含有4个操纵子,分别是ptnIPKC,cglDTEC,gcxKRCI和ibeRAT,共15个基因。该基因岛是由美国南加州大学医学院洛杉矶儿童医院黄胜和教授发现并命名,通过信息学预测该基因岛编码10
[Abstract]:Bacterial meningitis is the most common and serious infectious disease in central nervous system (central nerve). Despite the widespread use of antibiotics, the incidence of bacterial meningitis is between 5 and 40, while the incidence of neurological sequelae in survivors is as high as 30. One of the main causes of high infection and mortality in bacterial meningitis is that its pathogenesis is unclear. Escherichia coli E. coli is the most common gram-negative bacilli causing neonatal meningitis. Bacterial meningitis, most of which is known to occur in newborns, is the result of bacterial blood transmission. Entering the central nervous system is a necessary condition for the occurrence of bacterial meningitis. However, the mechanism of how the Escherichia coli in blood circulation penetrates the blood-brain barrier composed of (brain microvascular endothelial cells and BMECs, etc., remains to be studied. Some genes have been identified as related to bacterial penetration of the blood-brain barrier in meningitis Escherichia coli infection studies. TnphoA mutation assay showed that the gene ibeA played an important role in the process of bacterial invasion of BMEC and was unique to the virulence strain E.coli K1. The gene island containing ibeA was named GimA (a genetic island of meningitic E.coli containing ibeA). GimA). Its GC content (46.2%) was different from that of E.coli genome (50.8%). There are four operons, ptnIPKCU cglDTECgcxKRCI and ibeRAT. there are 15 genes. The gene island was discovered and named by Professor Huang Shenghe, Los Angeles Children's Hospital, School of Medicine, University of Southern California, USA. It is predicted by informatics that the gene island encodes 10.
