可控性心肌缺血动物模型的制作

【摘要】: 目的:建立小型猪可控性心肌缺血的动物模型。 材料与方法:健康成年小型猪10只,,雌雄对半。胸壁左侧第四肋间切口开胸安置自制水囊缩窄器,建立钝缘支可控性心肌缺血的动物模型。术后行缺血心电图试验、冠脉造影、微球技术检测水囊加压前后靶血管支配区局部心肌血流(CCBF)以及心肌HE染色观察靶血管支配区形态学变化。通过改进心电图导联方式、电极粘贴处皮肤处理办法以获取较为稳定的小型猪运动心电图。 结果:制作成功的8只小型猪在水囊加压后均可出现明显的缺血性心电图表现,同时冠脉造影显示钝缘支下游显影延迟、稀疏,钝缘支近乎完全闭塞,水囊压力撤除后心电图迅速恢复正常,钝缘支远端灌注恢复到水囊加压前状态。水囊加压前后靶血管支配区域CCBF明显减少(p<0.05)。HE染色显示靶血管支配区域心肌无梗死表现。用改进的心电图导联方式以及通过细致的皮肤预处理,我们获得了较为稳定的运动心电图。 结论:采用该自制水囊缩窄器制作的可控性心肌缺血动物模型,缺血效果确实、可靠,可控性强,且动物生存率高,优于以往报道结果。
[Abstract]:Objective: to establish an animal model of controllable myocardial ischemia in miniature pigs. Materials and methods: 10 healthy adult miniature pigs, half male and female. The fourth intercostal incision on the left side of the chest wall was used to establish an animal model of controlled myocardial ischemia with blunt marginal branch. After operation, ischemic electrocardiogram, coronary angiography and microsphere technique were used to detect the regional myocardial blood flow (CCBF) of the target vascular innervation area (CCBF) and the morphological changes of the target vascular innervation area by HE staining. The electrocardiogram (ECG) of miniature pig was obtained by improving the electrocardiogram (ECG) lead and treating the skin at the electrode. Results: all the 8 miniature pigs had obvious ischemic electrocardiogram (IECG) after water sac compression. Meanwhile, coronary angiography showed that the downstream development of the obtuse marginal branch was delayed, sparse, and the obtuse marginal branch was almost completely occluded. The electrocardiogram returned to normal quickly after the water capsule pressure was removed, and the distal perfusion of the blunt marginal branch returned to the state before the water sac pressure. CCBF in target area decreased significantly before and after water sac compression (p < 0.05). He staining showed that there was no myocardial infarction in target vascular innervation area. By using the improved electrocardiogram (ECG) lead and careful skin pretreatment, we obtained a stable exercise electrocardiogram (EKG). Conclusion: the animal model of controlled myocardial ischemia made by the self-made water capsule constrictor is effective, reliable and controllable, and the survival rate of the animal is high, which is better than that reported in the past.
