【摘要】:ALEX2基因(Arm proteins lost in epithelial cancers,X chromosome,2),属于ALEX家族成员,除ALEX2基因外,该家族还包括ALEX1和ALEX3基因。ALEX家族基因的共同特点是基因均定位于染色体Xq21.33-q22.2上;蛋白序列中都含有Armadillo repeat结构;蛋白序列高度同源,在蛋白N端都具有一个疏水的跨膜区。在组织表达分布上亦有很高的一致性:RT-PCR分析表明,在正常组织中除肝脏、胸腺组织低表达,外周血白细胞未见表达外均有广泛的表达;肿瘤组织中,在上皮组织来源的肿瘤组织(即癌组织)中低表达或不表达,提示ALEX家族蛋白在上皮组织来源的肿瘤的发生、发展过程中可能起重要作用,与癌组织的发生、恶化有潜在的关系。 Armadillo(arm)repeat结构首次发现于果蝇极性基因—armadillo,该基因在果蝇胚胎形成、维持上皮组织完整性和早期细胞极性形成中起重要作用。Arm repeat结构的特点是在真核和原核生物中都
[Abstract]:ALEX2 gene (Arm proteins lost in epithelial cancersX chromosome 2 (ALEX2) belongs to the Alex family. In addition to the ALEX2 gene, the common feature of this family is that the ALEX1 and ALEX3 genes are located on chromosome Xq21.33-q22.2, and the protein sequences contain Armadillo repeat structure. The protein sequence is highly homologous and has a hydrophobic transmembrane region at the N-terminal of the protein. There was also a very high consistency in the tissue expression distribution. The results of RT-PCR analysis showed that in normal tissues, the expression was low in liver and thymus tissues, but not in peripheral white blood cells, while in tumor tissues, there was no expression in peripheral white blood cells. The low expression or non-expression of Alex family proteins in tumour tissues derived from epithelial tissues may play an important role in tumorigenesis and development of epithelial tissues, and may play an important role in carcinogenesis.
