人巨细胞病毒感染致动脉粥样硬化的分子机制的研究

【摘要】:第一部分 人巨细胞病毒感染对人脐静脉内皮细胞黏附分子和RAGE受体表达的影响 目的 观察人巨细胞病毒(Human cytomegalovirus,HCMV)感染对人脐静脉内皮细胞(Human umbilical vein endothelial cells,HUVEC)的细胞间黏附分子-1(Intercellular adhesion molecule-1,ICAM-1)、血管细胞黏附分子-1(Vascular cell adhesion molecule-1,VCAM-1)及糖基化终产物受体(the Receptor for advanced glycation end products,RAGE)时序性表达的影响,研究HCMV感染致动脉粥样硬化(Atherosclerosis,AS)的作用机制。 方法 用HCMV感染HUVEC,采用RT-PCR方法检测不同感染时段细胞ICAM-1、VCAM-1及RAGE的mRNA表达。 结果 1.HCMV感染对HUVEC ICAM-1mRNA表达的影响 HCMV感染HUVEC细胞后,0h时ICAM-1mRNA有基础水平的表达,4h开始升高,8h表达量达高峰,12h开始回落,48h表达量仍维持在较高的水平,各时段与0h比较,均有统计学意义(P<0.01)。 2.HCMV感染对HUVEC VCAM-1mRNA表达的影响 HCMV感染HUVEC细胞后,0h时VCAM-1mRNA有基础水平的表达,4h开始升高,8h时达高峰,12h开始回落,24h回落更明显,,与0h比较,均有统计学意义(P<0.01),48h接近0h表达水平(P>0.05)。
