正常机体自身反应性B细胞的分布、编码基因及功能研究

【摘要】:在过去的10余年时间里,人们对 “自身免疫”的认识日趋深入。研究发现,自身反应性细胞不仅存在于自身免疫病患者,也存在于正常机体;应用某些自身抗原免疫,通过增强或扩大某些自身免疫反应,对一些疾病(自身免疫病、损伤修复)具有独特的治疗作用。这表明,“自身免疫”并不等同于“自身免疫病”,源自生理状态下的某些自身免疫反应可能对机体发挥着有益的作用。 以往研究表明:作为生理性自身免疫反应的组成成分,自身反应性B细胞存在于正常机体,并可分泌天然自身抗体(natural autoantibody,NAA),对维持机体自稳、介导免疫耐受的形成有重要作用。然而,其介导生理性自身免疫反应的机制尚不明确。在以往的研究基础上,本课题以角蛋白反应性B细胞及角蛋白反应性NAA为研究切入点,对其介导免疫耐受的机制进行探讨。 目的:分析正常机体角蛋白反应性B细胞的分布及比例,明确生理性自身免疫反应的存在;阐明其可变区基因编码特性,为进一步探讨角蛋白反应性B细胞的作用奠定基础;研究角蛋白反应性B细胞的抗原提呈作用,以及相应NAA在凋亡细胞清除中作用,探讨其介导免疫耐受的作用机制。 方法:实验内容共分5部分,实验方法简述如下: 1.检测自身反应性B细胞在正常机体的分布及比例 我们应用FITC对5种蛋白抗原进行了标记,用流式细胞仪(flow cytometry,
[Abstract]:In the past 10 years, people's understanding of "autoimmune" is getting deeper and deeper. It has been found that its own reactive cells exist not only in the autoimmune disease patients but also in the normal body. Some autoantigens are used and some autoimmune reactions are enhanced or expanded, and some diseases (autoimmune diseases, injury repair) are enhanced or expanded. It has a unique therapeutic effect. This shows that "autoimmune" is not equal to "autoimmune disease", and some autoimmune reactions from physiological states may play a useful role in the body.
Previous studies have shown that as a component of the physiological autoimmune reaction, self reactive B cells exist in the normal body, and can secrete natural autoantibodies (natural autoantibody, NAA), which plays an important role in maintaining the body's self stability and mediating the formation of immune tolerance. However, the mechanism of its mediated autoimmune reaction is not yet clear. On the basis of previous studies, this topic is based on the study of keratin reactive B cells and keratin reactive NAA as a breakthrough point, and the mechanism of its mediated immune tolerance is discussed.
Objective: to analyze the distribution and proportion of keratin reactive B cells in normal body, to clarify the existence of physiological autoimmune reaction, to clarify the coding characteristics of its variable region gene, to lay the foundation for further exploration of the role of keratin reactive B cells, to study the antigen presenting effect of keratin reactive B cells and the corresponding NAA in apoptotic cells. To explore the mechanism of action mediated immune tolerance.
Methods: the experiment is divided into 5 parts. The experimental methods are summarized as follows:
1. detect the distribution and proportion of reactive B cells in the normal body.
We used FITC to label 5 kinds of protein antigens, using flow cytometry (flow cytometry).
