大鼠纹状体发育过程中同源框基因islet-1的表达

【摘要】:纹状体是基底神经节接受传入信息的主要核团,在中枢神经系统有着不可替代的功能。而同源框基因由于其在生物进化过程中保持了相对的保守性,并且在神经系统有极为广泛的表达而成为近年来神经科学领域研究的热门之一。本实验针对LIM同源框基因家族的一员islet-1基因进行了研究,利用常规组织学染色和免疫组织化学染色等技术观察了胚胎第13天至成年大鼠纹状体发育过程中islet-1基因的表达状况。 结果显示:胚胎第13天,大鼠腹侧端脑已存在islet-1蛋白免疫组织化学反应阳性细胞;在不同发育时期,纹状体中islet-1阳性细胞的数量存在明显差异;同一发育阶段中,不同脑区该基因的表达状况亦不相同。 提示:中枢神经系统发育过程中islet-1基因的表达有显著的时间和空间差异;纹状体发育早期,islet-1蛋白阳性细胞广泛的分布说明该基因很可能表达于不同类型神经元前体细胞;随着纹状体逐渐发育成熟,islet-1基因的表达逐渐局限于少数神经元中,并可能对神经元的表型具备调节作用。
[Abstract]:Striatum is the main nucleus of basal ganglion which receives afferent information and has irreplaceable function in the central nervous system. The homobox gene has become one of the hot research fields in the field of neuroscience in recent years due to its relatively conservative nature in biological evolution and its wide expression in the nervous system. In this study, a member of islet-1 gene of LIM homobox gene family was studied. The expression of islet-1 gene during the development of striatum from the 13th day of embryo to adult rat was observed by routine histological staining and immunohistochemical staining. The results showed that there were islet-1 protein immunoreactive cells in the ventral cerebrum of rats on the 13th day of embryo, and there were significant differences in the number of islet-1 positive cells in striatum at different developmental stages. The expression of the gene was also different in different brain regions. The results suggest that there are significant temporal and spatial differences in the expression of islet-1 gene during the development of the central nervous system, and the wide distribution of islet-1 protein positive cells in the early stage of striatum development suggests that the gene may be expressed in different types of neuronal precursor cells. As the striatum matures, the expression of islet-1 gene is restricted to a few neurons and may regulate the phenotype of neurons.
