大鼠肝缺血再灌注损伤对卵巢机能代谢及内分泌的影响

【摘要】:目的:观察大鼠肝缺血再灌注(I/R)损伤过程中卵巢组织iNOS、MPO、MDA的变化及卵巢组织形态学改变,探讨卵巢组织能量代谢的变化,内分泌变化及其可能的机制。方法:建立肝缺血再灌注损伤动物模型,选健康Wistar大鼠48只,,均为雌鼠,随机分为对照组、缺血30min组(I组)、缺血30min即刻再灌注组(I/R组)、缺血30min再灌注后2h组(I/R2h组)、缺血30min再灌注后4h组(I/R4h组)、缺血30min再灌注后6h组(I/R6h组),每组8只。应用免疫组化法分别检测诱导型一氧化氮合酶(iNOS)的表达情况,同时行光镜检查,检测卵巢组织匀浆中髓过氧化物酶(MPO)、丙二醛(MDA)含量的变化,测定卵巢组织匀浆中ATP酶、乳酸脱氢酶(LDH)的活性及乳酸(LD)的含量,放免法检测血清雌二醇(E2)、孕酮(p)含量。结果:在肝缺血再灌注损伤过程中,iNOS在卵巢组织中的表达在I/R、I/R2h组与对照组、I组、I/R4h、I/R6h组比较差异有统计学意义(P<0.01)。光镜下I/R、IR2h组间质轻度水肿,少许炎细胞浸润,血管扩张充血,卵泡内卵丘颗粒细胞轻度变性。I/R4h、I/R6h组间质重度水肿,炎细胞浸润明显,大量血管扩张充血,卵泡内卵丘颗粒细胞明显变性。MPO在卵巢组织中的含量I、I/R6h组与对照组、I/R、I/R2h组比较差异有统计学意义(P<0.01,P<0.05),MDA在卵巢组织中的含量I、I/R2h、I/R4h、I/R6h组与对照组比较差异有统计学意义(P<0.01,P<0.05),I/R、I/R4h、I/R6h组与I组比较差异有统计学意义(P<0.01,P<0.05),I/R4h、I/R6h组与I/R组、I/R2h组比较差异
[Abstract]:Aim: to observe the changes of iNOS,MPO,MDA and morphology of ovarian tissue during hepatic ischemia reperfusion (I / R) injury in rats, and to investigate the changes of energy metabolism, endocrine and its possible mechanism. Methods: an animal model of hepatic ischemia-reperfusion injury was established. 48 healthy Wistar rats, all female rats, were randomly divided into control group. There were 8 rats in each group: 30min group (group I), immediate reperfusion group (group I / R), I/R2h group (group 2), I/R4h group (group I/R4h) and I/R6h group (group I/R6h). The expression of inducible nitric oxide synthase (iNOS) was detected by immunohistochemistry, and the changes of myeloperoxidase (MPO),) malondialdehyde (MDA) (MDA) in ovarian homogenate were detected by light microscopy, and ATP enzyme in ovarian homogenate was measured. The activity of lactate dehydrogenase (LDH) and the content of lactate (LD) were determined by radioimmunoassay. Serum estradiol (E2) and progesterone (p) were detected by radioimmunoassay. Results: there was significant difference in the expression of iNOS in ovarian tissues between the I / R / R 2 h group and the control group (P < 0.01) during hepatic ischemia-reperfusion injury (P < 0.01). Under light microscope, I / R IR2h group had mild edema, a little inflammatory cell infiltration, vasodilation, hyperemia, mild degeneration of cumulus granulosa cells in follicles, severe interstitial edema, obvious inflammatory cell infiltration, and massive vascular dilatation and hyperemia. Significant degeneration of cumulus granulosa cells in follicles. MPO content in ovarian tissue. There was significant difference in the content of MDA in ovarian tissues between the I / R / R 6h group and the control group (P < 0.01 P < 0.05). The content of MDA in the ovarian tissue was significantly different between the I / R _ 2 / R _ (6h) group and the control group (P < 0.01 / P < 0.05), and the content of MDA in the ovarian tissue was higher than that in the control group (P < 0.05). There was significant difference between the control group (P < 0.01) and I / R _ (4) H / R _ (6 h) group (P < 0.01 P < 0.05). There was a significant difference between the I / R _ 4 / R _ 4 / R _ (6 h) group and the I _ (R) / R _ (2) h group (P < 0.01 P < 0.05).
