动情周期对束缚应激大鼠皮质醇及脑内c-fos表达的影响

【摘要】: 为了研究动情周期不同时期对束缚应激的中枢整合的影响,在雌性大鼠动情周期的不同阶段给予1小时束缚应激。应用免疫组织化学法观测雌性大鼠脑不同区域c-fos蛋白表达的变化,应用放射免疫分析法检测血中皮质酮、雌二醇、孕酮的变化。结果:在1小时束缚应激组与对照组大鼠,均发现雌二醇在动情前期明显高于动情期与动情间期水平,而在动情周期的各期内,雌二醇水平在1小时束缚应激组与对照组无明显差别。孕酮在各组大鼠动情间期明显高于动情前期与动情期水平,在动情周期各期内,应激组与对照组无明显差别。皮质醇水平在对照组大鼠表现为动情前期高于动情期和动情间期,在各应激组大鼠均表现出较对照组明显增高,但在动情前期增高更明显。应激组大鼠在皮质、海马、缰核c-fos表达均较对照组增强;在海马的CA1、CA2、CA3区与额前皮质、犁状皮质、缰核外侧部c-fos表达在动情前期低于动情期和动情间期水平。而缰核的内侧c-fos表达与对照组相比明显增高,但在动情周期各期没有差别。实验结果:动情周期对束缚应激的中枢整合是有影响的,其中涉及到脑内的一些核团,包括海马、皮质,尤其是以前未提及的缰核,在应激反应中起很大作用。从而对女性的一些情感疾病如抑郁症、创伤后精神障碍等提供临床依据。
[Abstract]:In order to study the effect of estrus cycle on the central integration of restraint stress in different stages of estrus cycle, female rats were given 1 hour restraint stress at different stages of estrus cycle. The changes of c-fos protein expression in different regions of brain of female rats were observed by immunohistochemical method. The changes of corticosterone, estradiol and progesterone in blood were detected by radioimmunoassay. Results: in 1 hour restraint stress group and control group, estradiol was found to be significantly higher in early estrus than in estrus and estrus, but in each stage of estrus cycle. There was no significant difference in estradiol level between the 1 hour restraint stress group and the control group. The level of progesterone in estrous interphase was significantly higher than that in preestrus and estrus, but there was no significant difference between stress group and control group in estrous cycle. The level of cortisol in the control group was higher than that in the estrus phase and estrus interphase in the control group. The level of cortisol in the stress group was significantly higher than that in the control group, but the increase was more obvious in the preestrus phase. The expression of c-fos in cortex, hippocampus and habenula in stress group was higher than that in control group, and c-fos expression in prefrontal cortex, plough cortex and lateral habenular nucleus was lower than that in estrus and estrus interphase in the preestrus of hippocampus and prefrontal cortex, plow cortex and lateral part of habenula. The expression of medial c-fos in habenular nucleus was significantly higher than that in control group, but there was no difference in each stage of estrus cycle. Results: the estrus cycle has an effect on the central integration of restraint stress, which involves some nuclei in the brain, including hippocampus and cortex, especially habenular nucleus, which has not been mentioned before, and plays an important role in stress response. Therefore, some emotional diseases such as depression, post-traumatic disorder and so on provide clinical basis for women.
