老龄去势骨质疏松动物模型生物力学—生物化学—生物流变学研究

【摘要】: 随着社会的进步,对骨质疏松症的预防和治疗越来越得到重视,国内外学者对预防和治疗骨质疏松做了大量的研究,但是研究去势骨质疏松以中药、西药、电磁针、VD、雌激素防治骨质疏松骨骼的骨密度,骨骼的拉伸、压缩、弯曲、扭转、剪切、冲击力学性质和应力松弛、蠕变粘弹性(流变)性质。及研究去势骨质疏松以中药、西药等治疗动物血液的血磷、血钙、碱性磷酸酶指标、动物骨组织的显微结构改变情况未见报道。 鉴于相关研究和临床需要,作者设计了一组实验,将350只雌性大鼠随机分成正常对照组、模型组、中药治疗组、西药治疗组、电磁针治疗组、雌激素治疗组、VD治疗组。各组大鼠饲养第十五周周末在腹主动脉采血,取血清用于血磷、血钙和碱性磷酸酶测定,之后处死大鼠。取各组大鼠L3、L4、L5椎骨测量骨密度,取各组大鼠股骨粗隆部进行组织学观察;取各组大鼠骨进行拉伸实验;取各组大鼠骨进行压缩实验;取各组大鼠骨进行弯曲实验;取各组大鼠骨进行扭转实验;取各组大鼠骨进行剪切实验;取各组大鼠骨进行冲击实验;取各组大鼠骨进行应力松弛实验;取各组大鼠骨进行蠕变实验。 得出了各组大鼠骨密度、血清中血磷、血磷、碱性磷酸酶等生化指标。得出了各组大鼠骨显微结构分析结果。得出了各组大鼠骨拉伸、压缩、弯曲、应力、应变、弹性模量等数据。得出了各组大鼠骨扭转剪应力、扭矩、扭转角数据,得出各组大鼠骨骼冲击功、冲击韧性数据。得出了各组大鼠骨骼5400S应力松弛量、蠕变量,得出了各组大鼠骨骼归一化应力松弛函数,归一化蠕变函数,以期为骨质疏松的研究和临床提供理论基础。
[Abstract]:With the progress of society, more and more attention has been paid to the prevention and treatment of osteoporosis. Scholars at home and abroad have done a lot of research on the prevention and treatment of osteoporosis, but the study of ovariectomized osteoporosis by Chinese medicine, western medicine, electromagnetic acupuncture, VD, Estrogen prevents bone mineral density (BMD), bone stretching, compression, bending, torsion, shear, impact and stress relaxation, creep viscoelastic (rheological) properties. There was no report on the changes of blood phosphorus, calcium, alkaline phosphatase and bone microstructure of ovariectomized osteoporosis treated with traditional Chinese medicine and western medicine. In view of the relevant research and clinical needs, the author designed an experiment in which 350 female rats were randomly divided into normal control group, model group, traditional Chinese medicine treatment group, western medicine treatment group, electromagnetic acupuncture group, estrogen treatment group and VD treatment group. Blood samples were collected from abdominal aorta at the end of the fifteenth week of feeding in each group. Serum was collected for determination of serum phosphorus, calcium and alkaline phosphatase, and then the rats were killed. The bone mineral density (BMD) was measured by L3, L4, L5 vertebrae, the femoral trochanter of each group was observed histologically, the bone of each group was taken for stretching test, the bone of each group was compressed, the bone of each group was obtained for bending test. Every group of rat bone for torsion test; each group of rat bone for shear test; each group of rat bone for impact test; each group of rat bone for stress relaxation test; each group of rat bone for creep test. The biochemical indexes of bone mineral density, serum phosphorus and alkaline phosphatase were obtained. The results of bone microstructure analysis in each group were obtained. The data of bone stretching, compression, bending, stress, strain and elastic modulus were obtained. The data of bone torsion shear stress, torque and torsion angle were obtained, and the data of bone impact work and impact toughness were obtained. The stress relaxation quantity and creep amount of rat bone 5400S in each group were obtained. The normalized stress relaxation function and the normalized creep function were obtained in each group of rats in order to provide a theoretical basis for the study and clinical application of osteoporosis.
