TGFβ1介导新生大鼠心肌成纤维细胞Ⅰ型和Ⅳ型胶原合成的研究

【摘要】: 背景:心肌纤维化系心肌成纤维细胞增殖、细胞外基质合成及细胞凋亡等一系列复杂因素调控所产生的结果。在心肌纤维化的进程中,转化生长因子-β_1(TGF-β_1)起着重要调控作用,它通过加速心肌成纤维细胞的分化、抑制心肌成纤维的凋亡、促进细胞外基质的产生而刺激纤维化反应。这些信号分子的作用受细胞的内环境的影响,且具有器官特异性,细胞与细胞之间以及细胞与基质问的相互作用也能促进或阻止该信号通路。迄今为止,以TGF-β_1干预心肌成纤维细胞的体外研究中,心肌成纤维细胞的生物学反应程度及其作用机制尚不清楚。因此,以TGF-β_1干预心肌成纤维细胞,通过检测其下游通路的信号分子,,来探讨其在心肌纤维化中的作用,对于改善心脏疾病的治疗及降低疾病的纤维化所致的死亡具有重要意义。 目的:观察TGF-β_1对原代培养的新生大鼠心肌成纤维细胞Ⅰ型与Ⅳ胶原表达的影响,探讨TGF-β_1在心肌纤维化发生发展中的可能作用机制。 方法:取出生3-7天的SD大鼠心脏,利用酶消化法结合差速贴壁体外培养心肌成纤维细胞;分别用不同浓度的TGF-β_1(TGFβ1=0ng/ml、1ng/ml、10ng/ml)干预心肌成纤维细胞,分别于干预后5h、24h,采用RT-PCR及免疫细胞化学染色法检测Ⅰ型胶原和Ⅳ型胶原的表达。 结果: 1、用胰酶与Ⅱ型胶原酶合用消化分离新生大鼠心肌成纤维细胞,可将心肌细胞和心肌成纤维细胞分离,心肌成纤维细胞抗波形蛋白(vimentin)染色阳性,抗α-横纹肌肌动蛋白(α-sarcomeric actin)染色阴性。 2、用0ng/ml(对照组),1ng/ml和10ng/ml的TGF-β_1干预心肌成纤维细胞5h,RT-PCR检测Ⅰ型胶原/β-actin光密度值分别为0.61±0.02,0.73±0.03和0.86±0.04,光密度值随着TGFβ1浓度的增加而增加,各组比较,差异有显著性(P<0.01);干预心肌成纤维细胞24h后,RT-PCR检测Ⅰ型胶原/β-actin光密度值分别为0.65±0.03,0.91±0.02和0.98±0.02,光密度值随着TGF-β_1浓度的增加而增加,各组比较,差异有显著性(p<0.01)。 3、用0ng/ml(对照组),1ng/ml and 10ng/ml的TGF-β_1干预心肌成纤维细胞5h,RT-PCR检测Ⅳ型胶原/β-actin光密度值分别为0.58±0.06,0.71±0.02,0.87±0.02,光密度值随着TGF-β_1浓度的增加而增加,各组比较,差异有显著性(p<0.01);干预心肌成纤维细胞24h后,RT-PCR检测Ⅳ型胶原/β-actin光密度值分别为0.62±0.01,0.83±0.05,0.96±0.02,光密度值随着TGF-β_1浓度的增加而增加,各组比较,差异有显著性(p<0.01)。 4、用0ng/ml(对照组),1ng/ml and 10ng/ml的TGF-β_1干预心肌成纤维细胞5h,24h后,用免疫细胞化学染色法检测心肌成纤维细胞Ⅰ型胶原和Ⅳ型胶原的表达情况,结果显示,随着浓度的增加,胞浆颜色由浅黄色变成深黄色。 结论: 1、成功分离和培养新生大鼠心肌成纤维细胞。 2、TGF-β_1可使心肌成纤维细胞Ⅰ型胶原和Ⅳ型胶原表达上调,呈时间和剂量依赖性。 3、TGF-β_1能促进心肌成纤维细胞Ⅰ型胶原和Ⅳ型胶原的合成,提示TGF-β_1在心肌纤维化中可能起了重要作用。
