人疱疹病毒8型重组蛋白的制备和应用及其感染的流行病学调查

【摘要】:第一部分 山东地区部分HIV感染者及献血者人疱疹病毒8型感染的流行病学调查 目的 检测山东地区部分HIV(+)/KS(-)感染者及献血者血清或血浆中抗人疱疹病毒8型(HHV-8)IgG抗体的存在情况,并进一步对抗HHV-8阳性标本中HHV-8 DNA的存在情况进行检测;从而了解HHV-8在我国山东地区HIV感染者及献血者中的感染情况,并探讨HHV-8经输血传播的可能性。 方法 86份HIV(+)/KS(-)血清(来源于山东地区HIV感染者)及520份献血者血浆(主要来源于济南地区)中抗HHV-8 IgG抗体的检测采用ELISA方法;抗HHV-8阳性标本(血清或血浆)中HHV-8 DNA的检测采用实时荧光定量PCR技术(SYBR Green Ⅰ荧光染料法)。 结果 1 血清学检测结果 1.1 86份HIV(+)/KS(-)血清中抗HHV-8抗体检出率为16.275%(14/86),与对照组(健康献血者血浆)比较有显著性差异(P<0.05)。 1.2 520份献血者血浆中抗HHV-8抗体检出率为5.962%(31/520),其中男性献血者血浆中抗HHV-8 IgG抗体的检出率为6.367%(17/120),女性献血者血浆中抗HHV-8 IgG抗体的检出率为5.534%(14/110)。统计学分析表明,不同性别、年龄献血者血浆中抗HHV-8 IgG抗体的检出率无显著性差异(P>0.05)。 1.3 HBsAg(+)、抗HCV(+)以及抗梅毒螺旋体(+)献血者血浆中抗HHV-8 IgG抗体的检出率分别为4.065%、4.651%及6.579%,统计学分析表明,,抗HHV-8抗体的检出与HBsAg、抗HCV以及抗梅毒螺旋体的检出相关性不大(P>0.05)。 2 PCR检测结果 所有抗HHV-8阳性标本(血清或血浆)中均未检出HHV-8 DNA的存在。
