慢性阻塞性肺疾病寒饮蕴肺证大鼠模型的建立

【摘要】: 本文运用传统理论研究与现代实验研究相结合的方法,对“慢性阻塞性肺疾病(COPD)寒饮蕴肺证”进行了深入的理论探讨和严谨的实验研究,旨在揭示寒饮蕴肺证在具体疾病中的病变机理。在理论研究方面,主要探讨了肺阳在肺主行水中的作用,以及肺阳虚导致肺失宣降,肺主行水功能失调,水饮停聚于肺,水为阴邪,肺阳本虚,温煦不足,,从而水饮寒化,形成寒饮蕴肺之证。COPD作为呼吸系统的一种常见病,在中医学中属于咳嗽、喘证、肺胀等范畴,所以治疗COPD寒饮蕴肺证以温肺阳,补肺气,祛寒饮为主。 为深化对“寒饮蕴肺”的认识,进一步明确其微观机制,本文在理论探讨的基础上,运用当前病证结合的造模理论,结合中医传统病因和现代医学病理造模方法,成功复制了COPD寒饮蕴肺证大鼠模型。本模型的复制方法综合了病和证的研制方法,运用了与COPD寒饮蕴肺证的形成关系更为密切的多种因素,更加符合COPD寒饮蕴肺证形成的条件和病机。借助小动物肺功能仪、放免测定、免疫组化等技术,以多途径、多指标、同步观察的方法,对造模动物进行了实验观察及药物干预,在症状表现方面,基本符合该病证的临床表现,微观指标如肺组织病理形态学、肺功能、cAMP、cGMP等结果基本符合病COPD及以虚寒证为主的病证表现;用药干预组的检测指标有明显改善,从而证明了模型的可靠性。为肺藏象的现代研究及中医辨证的客观化提供了实验依据。
[Abstract]:Based on the combination of traditional theoretical research and modern experimental research, this paper makes a thorough theoretical study and rigorous experimental study on "(COPD) cold drink accumulation of lung syndrome" in chronic obstructive pulmonary disease (COPD). The purpose of this study was to reveal the pathological mechanism of cold-drink syndrome of lung accumulation in specific diseases. In the theoretical research, it mainly discusses the role of lung yang in the lung water, as well as the deficiency of lung yang leading to the loss of the lung, the imbalance of the function of the lung, the water drinking stops in the lung, the water is Yin evil, the deficiency of the lung yang is the deficiency of the lung yang, and the warmth is insufficient, so that the water drinks cold. COPD, as a common disease of respiratory system, belongs to the category of cough, asthma and lung distension in traditional Chinese medicine, so the treatment of COPD cold drink accumulation lung syndrome is mainly to warm the lung yang, tonify the lung qi, and dispel cold drink. In order to deepen the understanding of "cold drink to accumulate the lung" and to further clarify its microscopic mechanism, this paper, on the basis of theoretical discussion, applies the current model making theory of combining disease and syndrome, combining the traditional etiology of traditional Chinese medicine and modern medical pathological modeling method. The rat model of COPD cold drink accumulation of lung syndrome was successfully duplicated. The replication method of this model synthesizes the development method of disease and syndrome, and uses many factors which are more closely related to the formation of COPD cold drink accumulation lung syndrome, which is more in line with the conditions and pathogenesis of the formation of COPD cold drink accumulation lung syndrome. With the help of small animal lung function instrument, radioimmunoassay, immunohistochemistry and other techniques, the animal model was observed and drug intervention was carried out with the methods of multi-channel, multi-index and synchronous observation. The results of microscopic indexes such as lung histopathology, pulmonary function and cAMP,cGMP basically accord with the clinical manifestations of the disease and the symptoms of deficiency and cold syndrome. The test indexes of the intervention group were improved obviously, which proved the reliability of the model. It provides the experimental basis for the modern study of lung storage and the objectification of TCM syndrome differentiation.
