江西省2009年至2012年甲型H3N2流感病毒耐药性分析

【摘要】:目的分析江西省2009-2012年甲型H3N2流感病毒M2以及NA基因的特点,掌握其耐药情况,为临床治疗和疾病控制提供参考。方法从江西省流感监测网中随机选择19株甲型H3N2流感病毒,经核酸提取和one-step RT-PCR扩增M以及NA基因片段,双向序列测定,采用DNAStar5.0和Mage4.0序列分析软件分析M2以及NA基因特征以及耐药性位点。结果 19株毒株M2基因31位氨基酸均由丝氨酸(S)突变为天冬酰胺(N);所有毒株NA蛋白催化活性位点和辅助位点均未发生氨基酸替换。结论 19株毒株均对金刚烷胺类药物耐药,对流感病毒神经氨酸酶抑制剂药物均敏感,但仍应加强对流感病毒的耐药性监测。
[Abstract]:Objective to analyze the characteristics of M 2 and NA genes of influenza A virus (H3N2) in Jiangxi Province from 2009 to 2012, and to provide reference for clinical treatment and disease control. Methods 19 strains of influenza A virus (H3N2) were randomly selected from Jiangxi Influenza Surveillance Network. M and NA gene fragments were amplified by nucleic acid extraction and one-step RT-PCR and sequenced in two directions. The characteristics and drug resistance sites of M2 and NA genes were analyzed by DNAStar5.0 and Mage4.0 sequence analysis software. Results the amino acid position 31 of M2 gene of 19 strains was mutated from serine (S) to asparagine (N);. There were no amino acid substitutions in NA protein catalytic active sites and auxiliary sites of all strains. Conclusion all 19 strains are resistant to amantadine and sensitive to influenza virus neuraminidase inhibitors, but the surveillance of resistance to influenza virus should be strengthened.
