【摘要】:目的对比PCR-线性探针杂交酶显色法(简称线性探针法)与BACTEC MGIT960液体培养及药敏在结核分枝杆菌检出率的差异,评价两种方法检测结核分枝杆菌对利福平和异烟肼耐药性的应用效果。方法选取医院结核科2013年8月-2015年5月369例确诊及疑似结核病的住院患者送检标本,同时采用线性探针法和BACTEC MGIT960对其进行结核分枝杆菌及利福平和异烟肼耐药性检测,数据采用SPSS 13.0软件进行统计分析。结果线性探针法检测结核分枝杆菌对利福平耐药性的灵敏度为90.4%、特异度为97.3%、Kappa值为0.872;对异烟肼耐药性检测的灵敏度为86.6%、特异度为96.3%、Kappa值为0.841,两种方法在利福平和异烟肼耐药性检测中具有较好的一致性。结论 PCR-线性探针杂交酶显色法是一种快速、准确的结核分枝杆菌及其对利福平和异烟肼耐药性的诊断方法,对耐多药结核病的快速诊断具有显著的优点。
[Abstract]:Objective to compare the PCR- linear probe hybridization enzyme colorimetric method (the linear probe) the difference in detection rate and BACTEC MGIT960 liquid culture and drug resistance in Mycobacterium tuberculosis, to evaluate the application effect of two methods for detection of Mycobacterium tuberculosis resistance to rifampin and isoniazid. Methods selected the hospital department of tuberculosis in August 2013 -2015 year in May 369 cases and the samples were hospitalized with suspected tuberculosis patients, at the same time using a linear probe and BACTEC MGIT960 for detection of Mycobacterium tuberculosis resistance to rifampin and isoniazid and the SPSS 13 software was used for statistical analysis. The results of detection of Mycobacterium tuberculosis rifampin resistance probe method of linear sensitivity was 90.4%, specificity was 97.3%, the Kappa value is 0.872 the sensitivity to isoniazid resistance; detection was 86.6%, specificity was 96.3%, the Kappa value is 0.841, two kinds of methods in rifampin and isoniazid Conclusion: PCR- linear probe hybridization enzyme chromogenic assay is a rapid and accurate method for the diagnosis of Mycobacterium tuberculosis and its resistance to rifampicin and isoniazid, and has a significant advantage for the rapid diagnosis of multidrug-resistant tuberculosis.
