利福喷丁对大鼠细胞因子及巨噬细胞吞噬功能的影响

【摘要】:目的:研究利福喷丁对大鼠细胞因子IFN-γ、IL-12、IL-6及巨噬细胞吞噬功能的影响。方法:1.60只SD雄性大鼠,二月龄,体重为180±30g,随机分为实验组和对照组,每组为30只,每组再随机分为喂药一月组(A组)、喂药三月组(B组)、喂药三月停喂药一月组(以下简称为停药一月组)(C组)(每组为10只)。2.实验组用利福喷丁灌胃,对照组用0.1%羧甲基纤维素钠液灌胃,其余喂养条件相同。3.ELISA法检测大鼠血清IFN-γ、IL-12、IL-6含量,琼脂平板法检测大鼠血清溶菌酶含量,硝酸铅法显示大鼠腹腔巨噬细胞内酸性磷酸酶活性。IFN-γ、IL-12、IL-6、溶菌酶结果用t检验统计;酸性磷酸酶用轶和检验统计。结果:1.实验组IFN-γ在喂药三月较对照组有明显降低,差异有统计学意义(P<0.05),在喂药一月、停药一月较对照组比较,差异无统计学意义;实验组IL-12在喂药一月、喂药三月、停药一月较对照组比较,差异均无统计学意义;实验组溶菌酶在喂药三月较对照组有明显升高,差异有统计学意义(P<0.05),在喂药一月、停药一月较对照组比较,差异均无统计学意义;实验组酸性磷酸酶在喂药一月、喂药三月较对照组有明显升高,差异均有统计学意义(P<0.05),在停药一月较对照组比较,差异无统计学意义。2.对照组组内比较:喂药三月组、停药一月组溶菌酶较喂药一月组比较,结果有明显降低,差异均有统计学意义(P<0.05);喂药三月组、停药一月组酸性磷酸酶较喂药一月组比较,结果有明显升高,差异均有统计学意义(P<0.05)。IFN-γ、IL-12在对照组内比较,差异均无统计学意义。3.ELISA法在所有组中均未检测出指标IL-6,可能的原因是IL-6在大鼠体内的含量很低,未达到ELISA检测的最低检测标准。结论:1.利福喷丁显著降低大鼠血清IFN-γ的含量。利福喷丁降低大鼠血清IL-12含量,但降低未达到统计学差异。利福喷丁显著提高大鼠血清溶菌酶含量,并且显著增强大鼠腹腔巨噬细胞酸性磷酸酶活性。2.随着大鼠月龄的增长,大鼠血清溶菌酶含量降低,腹腔巨噬细胞酸性磷酸酶活性增强。3.停药后,大鼠血清IFN-γ、IL-12和溶菌酶含量恢复,,大鼠腹腔巨噬细胞酸性磷酸酶活性恢复。
[Abstract]:Objective: to study the effect of rifapentine on cytokine IFN- 纬 -IL-12 IL-6 and macrophage phagocytosis in rats. Methods: one hundred and sixty SD male rats, aged two months, with body weight of 180 卤30 g, were randomly divided into experimental group and control group with 30 rats in each group. Each group was randomly divided into three groups: group A (group A), group B (group B), group A (group A), group B (group B), group B (group B), group C (group C), group C (10 rats in each group). In the control group, 0.1% sodium carboxymethylcellulose was used to gavage, and the other feeding conditions were the same. 3. Elisa method was used to detect the content of serum IFN- 纬 -IL-12 and IL-6, and Agar plate method was used to detect the content of serum lysozyme in rats. Lead nitrate method showed that the activity of acidic phosphatase in peritoneal macrophages. IFN- 纬 -IL-12 and IL-6, lysozyme results were counted by t test, acid phosphatase by Yi and test statistics. Results: 1. IFN- 纬 in experimental group was significantly lower than that in control group in three months. The difference was statistically significant (P < 0.05). There was no significant difference in IL-12 between the control group and the control group in one month and one month after the drug was given, but there was no significant difference between the experimental group and the control group in one month, three months, and one month, respectively. The lysozyme of the experimental group was significantly higher than that of the control group in three months, the difference was statistically significant (P < 0.05), but there was no significant difference between the experimental group and the control group in one month, and the acid phosphatase in the experimental group was fed for one month. Compared with the control group, the difference was statistically significant (P < 0.05). There was no significant difference between the control group and the control group in one month. The comparison between the control group and the control group: the lysozyme in the three-month group and the one-month group were significantly higher than those in the one-month group. The results showed that the difference was statistically significant (P < 0.05), and the acid phosphatase level was significantly higher in the three months group than that in the control group (P < 0.05. IFN- 纬 -IL-12, P < 0.05, P < 0.05, P < 0.05, P < 0.05), and the level of IL-12 in the control group was significantly higher than that in the control group (P < 0.05, P < 0.05). There was no significant difference. 3. Elisa did not detect IL-6 in all groups, which may be due to the low content of IL-6 in rats. Conclusion: 1. Rifapentine can significantly decrease the content of IFN- 纬 in serum of rats, and rifapentine can decrease the content of IL-12 in serum of rats. But the decrease was not statistically significant. Rifapentine significantly increased the content of serum lysozyme and increased the activity of acid phosphatase in peritoneal macrophages of rats significantly. With the increase of the age of rats, the content of serum lysozyme decreased. The activity of acidic phosphatase in peritoneal macrophages was enhanced .3.After drug withdrawal, the contents of IL-12 and lysozyme in serum IFN- 纬 and lysozyme were recovered, and the activity of acid phosphatase in peritoneal macrophages was recovered.
