绿原酸、异绿原酸对烟曲霉菌生物被膜抑制作用的体外研究

【摘要】:目的:通过在体外复制烟曲霉菌(Aspergillus fumigates)的生物被膜(Biofilm, BF)模型,研究绿原酸(Chlorogenic acid,CRA)、异绿原酸(Isochlorogenic acid,IRC)对生物被膜的影响。 方法:选取临床分离的烟曲霉菌构建体外生物被膜模型,微量液基稀释法测定最低抑菌浓度(Minimal inhibition concentration, MIC),,将菌株分为空白对照组、实验组(CRA1024μg/ml、CRA512μg/ml、CRA256μg/ml、IRC1000μg/ml、IRC500μg/ml、IRC250μg/ml),扫描电子显微镜(Scanningelectron microscopy,SEM)、激光共聚焦扫描显微镜(Laser confocal scanningmicroscope, CLSM)定性观察BF;结晶紫染色法半定量生物被膜量。 结果:37℃培养24h烟曲霉菌形成早期BF,48h形成成熟期BF。实验组烟曲霉菌BF均较其相应空白对照组空洞、稀疏,但菌丝未见明显减少,死活菌数也无明显差别。CRA组早期结晶紫半定量结果分别为(1.05±0.19)、(1.14±0.26)、(1.16±0.14),IRC组为(1.18±0.06)、(1.21±0.06)、(1.26±0.04),均少于空白对照组(1.91±0.17),差异具有统计学意义(P0.05);CRA组成熟期结晶紫半定量结果为(2.25±0.05)、(2.27±0.05)、(2.31±0.03),IRC组为(2.26±0.02)、(2.27±0.02)、(2.29±0.04),也均少于空白对照组(2.36±0.01),差异具有统计学意义(P0.05)。 结论:1、可以在体外建立烟曲霉菌生物被膜模型;2、CRA、IRC对体外烟曲霉菌BF形成有抑制作用。
[Abstract]:Objective: to study the effect of Lv Yuan acid Chlorogenic acidoglycan (Isochlorogenic acidogenic) on biofilm by replicating the biofilm model of Aspergillus fumigatesin vitro. Methods: a biofilm model was established in vitro by clinical isolates of Aspergillus fumigates.Methods: the biofilm model of Aspergillus fumigatesin vitro was established. Minimal inhibition concentration was determined by microdilution method. The strains were divided into blank control group. CRA1024 渭 g / ml CRA512 渭 g / ml CRA256 渭 g / ml IRC1000 渭 g / ml IRC500 渭 g / ml IRC250 渭 g / ml, scanning electron microscope (SEM), laser confocal scanning microscope (laser confocal scanning microscope), laser confocal scanning microscope (laser confocal scanning microscope), laser scanning microscope (CLSM), and semi-quantitative biofilm quantity of CRA 256 渭 g / ml.
