组蛋白去乙酰化酶抑制剂激活HIV储存库的研究进展

【摘要】:即使是极早期接受规范化抗反转录病毒治疗(ART)的艾滋病病毒(HIV)感染者,HIV也会在CD+4T淋巴细胞中形成病毒储存库。储存库的形成与整合的前病毒的转录沉默相关,且至少部分由组蛋白去乙酰化酶(HDACs)所诱导。HDACs是一组染色质相关的蛋白质,可以调节组蛋白的乙酰化水平以及脱氧核糖核酸(DNA)与转录因子的亲和程度。值得注意的是,通过抑制HDACs的活性,可以有效地激活储存库中的HIV。这一发现衍生出"shock and kill"的设想,可能达到清除储存库的目的,也可能成为HIV功能性治愈的策略之一。
[Abstract]:Even for (HIV) infected with HIV who received standardized antiretroviral therapy for (ART) at an early stage, HIV could form a viral repository in CD 4 T lymphocytes. The formation of the repository is related to the transcriptional silencing of integrated proviruses, and is at least partially induced by histone deacetylase (HDACs). HDACs is a group of chromatin related proteins. It can regulate the acetylation level of histone and the affinity between (DNA) and transcription factor. It is worth noting that by inhibiting the activity of HDACs, the HIV. in the repository can be activated effectively This finding derives from the idea of "shock and kill", which may be used to clear the repository and become one of the strategies for functional cure of HIV.
