塞来昔布对细粒棘球蚴原头节生长及p-ERK2表达的影响

【摘要】:目的探索塞来昔布对细粒棘球蚴原头节体外生长及p-ERK2表达的影响。方法实验分组为1640培养基组、DMSO组、塞来昔布溶液(50、100、150、200μmol/L)中体外孵育,在倒置焦镜下观察原头节的形态变化,同时通过0.1%伊红染色显示原头节活力。运用Western blot检测DMSO、100、150、200μmol/L塞来昔布处理细粒棘球蚴原头节48h后p-ERK2蛋白的表达量。结果体外孵育7d后,DMSO组及空白1640对照组原头节的活力几乎没有改变。塞来昔布作用1d后,100、150、200μmol/L组原头节的活力均下降;作用3d后,200μmol/L组原头节全部被杀死,150μmol/L组的原头节活力为43.9%。100μmol/L、50μmol/L组对原头节的活力也有明显下降,但不及高浓度组显著。Western blot结果显示P-ERK2的表达量随浓度增加而下降趋势明显。结论塞来昔布能够抑制细粒棘球蚴原头节生长,这可能与塞来昔布下调P-ERK2的表达进而抑制ERK2信号传导通路有关。
[Abstract]:Objective to investigate the effect of celecoxib on the growth and p-ERK2 expression of Echinococcus granulosus in vitro. Methods the experimental groups were divided into 1640 medium group, DMSO group and celecoxib solution (50100150200 渭 mol / L). The morphological changes of the original head segment were observed under inverted focal microscope, and the activity of the original head segment was shown by 0.1% Ihong staining. The expression of p-ERK2 protein in the original head segment of Echinococcus granulosus treated with DMSO, 100150200 渭 mol / L celecoxib for 48 h was detected by Western blot. Results after incubation in vitro for 7 days, the activity of the original head node in DMSO group and blank 1640 control group was almost unchanged. After celecoxib treatment for 1 day, the activity of the original head decreased in 100150200 渭 mol / L group. After 3 days of treatment, all the primary head segments of 200 渭 mol / L group were killed, and the activity of the original head section of 150 渭 mol / L group was 43.9%. 100 渭 mol / L, and the activity of 50 渭 mol / L group also decreased significantly. However, it was not as significant as that of high concentration group. Western blot results showed that the expression of P-ERK2 decreased significantly with the increase of concentration. Conclusion celecoxib can inhibit the growth of the original head segment of Echinococcus granulosus, which may be related to the down-regulation of P-ERK2 expression by celecoxib and the inhibition of ERK2 signal transduction pathway.
