泡球蚴囊液对大鼠肝星状细胞增殖和凋亡影响

【摘要】:目的:探讨泡球蚴囊液对体外大鼠肝星状细胞(HSC-T6)增殖和凋亡影响的初步研究。 方法: HSC-T6细胞进行体外培养,用CCK-8检测并绘制HSC-T6的生长曲线、不同浓度泡球蚴囊液(0mg/ml-13.5mg/ml)干预24h时的增殖活性,并应用荧光倒置显微镜观察囊液作用前后的HSC-T6形态变化;采用流式细胞仪检测并分析HSC-T6细胞周期和凋亡情况。 结果:①不同浓度泡球蚴囊液(0mg/ml-13.5mg/ml)干预的24h后部分细胞固缩,,呈脱落前兆,而部分HSC-T6细胞收缩成长梭形,细胞生出许多细长的伪足。②浓度为0mg/ml-0.85mg/ml时则对HSC-T6细胞增殖活性有促进作用,浓度为0.85mg/ml-13.5mg/ml时则对HSC-T6细胞增殖活性有明显抑制作用,且都有时间和浓度依懒性。③流式细胞仪结果显示随着泡球蚴囊液浓度(0mg/ml-6.8mg/ml)的增加,大鼠HSC-T6细胞的凋亡率逐渐增加。 结论:在一定浓度范围内,泡球蚴囊液对体外培养大鼠肝星状细胞增殖有一定的促进作用,并可促进其凋亡;泡球蚴囊液能够促进肝星状细胞由G0/G1期进入S期。
[Abstract]:Aim: to investigate the effect of alveolar hydatid cyst fluid on proliferation and apoptosis of rat hepatic stellate cells (HSC-T6) in vitro. Methods: HSC-T6 cells were cultured in vitro, the growth curve of HSC-T6 was detected and drawn by CCK-8, the proliferation activity of different concentrations of hydatid cyst fluid (0mg/ml-13.5mg/ml) for 24 hours was observed, the morphological changes of HSC-T6 were observed by fluorescence inverted microscope, and the cell cycle and apoptosis of HSC-T6 cells were detected and analyzed by flow cytometry. Results: 1 after 24 hours of intervention with different concentrations of alveolar hydatid cyst fluid (0mg/ml-13.5mg/ml), some cells shrank and showed exfoliative precursors, while some HSC-T6 cells contracted and grew fusiform, and the cells produced many slender pseudopods. 2 the concentration of 0mg/ml-0.85mg/ml promoted the proliferation activity of HSC-T6 cells. At the concentration of 0.85mg/ml-13.5mg/ml, the proliferation activity of HSC-T6 cells was significantly inhibited, and both time and concentration were lazy. 3 the results of flow cytometry showed that the apoptosis rate of rat HSC-T6 cells increased gradually with the increase of the concentration of alveolar hydatid cyst fluid (0mg/ml-6.8mg/ml). Conclusion: in a certain concentration range, the alveolar hydatid cyst fluid can promote the proliferation and apoptosis of rat hepatic stellate cells in vitro, and the alveolar hydatid cyst fluid can promote the hepatic stellate cells from G0/G1 phase to S phase.
