HLA-DP/HLA-DQ基因多态性与乙肝病毒感染及进展易感性的关联研究

  乙肝病毒感染及其进展的疾病是常见而且影响重大的感染性疾病之一,目前全球约2亿4千万人群为慢性乙肝病毒感染,目前中国不少地区仍为乙肝高流行区,每年因乙肝感染病情加重死亡的患者多达5万人。影响乙肝病毒感染因素中,宿主遗传特性被发现对乙肝病毒感染及进展有重要影响。近年来发表的几个全基因组关联研究发现了位于6号染色体HLA基因家族上的单核苷酸多态性位点(SN P)与乙肝病毒感染及感染后疾病进展密切相关。HLA基因家族富集了多个免疫相关蛋白的编码区,我们将通过对这一区域(主要是HLA-DP/DQ基因簇)的精细定位来发现更多的影响乙肝病毒感染的基因位点,研究这些位点的作用模式、相互影响以及对感染后疾病进展的影响。方法:我们在本研究中收集了1440例健康入和1648例乙肝感染者的样本,分别提取DNA后使用zju-iselect定制芯片对HLA基因区域进行了高密度的SNP位点基因分型精细定位。之后对芯片数据结果分别用PLINK软件和Haploview软件做单个SNP位点和各连锁区块(linkage block, LD)上单倍型的关联分析以及条件关联分析,另外还使用R语言程序包haplo.stat s作...
