柯尔克孜羊超数排卵、同期发情及胚胎移植效果研究

【摘要】:无论是对绵羊的超数排卵和同期发情,还是对绵羊的胚胎移植,虽然国内外的诸多学者已经研究了数十年,但在各种各样的因素影响之下,取得的效果并不理想。在本研究中,以柯尔克孜绵羊作为研究对象,不仅详细地分析了影响该种绵羊超排效果和同期发情的主要因素,而且深入地分析了影响其胚胎移植妊娠效果的关键因素,初步建立高寒牧区绵羊的胚胎移植技术体系。试验结果表明:像阿合奇县等高寒牧区进行绵羊超数排卵最佳时间为9月一10月中旬,但进入12月份后超排效果出现不太稳定的趋势;通过应用PG+FSH法和CIDR+FSH法对柯尔克孜羊进行超排处理,即使平均的采胚数不存在显著的差异,不过通过第2种方法计算出来的平均可用胚数出现一定的减小;在激素总量相同的情况下,从药物成本来看,对柯尔克孜羊应用PG十FSH法超排的效果优于CIDR十FSH法;在每组的柯尔克孜羊中,都通过药物进行作用,用量可以有210IU、250IU、270IU、320IU的FSH,实现超排的处理能够发现,每只作用250IU得到的效果最好,平均的采胚数和可用的胚数,都比其他组别更加显著(P0.01);对柯尔克孜羊首次超排后、间隔1个和2个情期超排的排卵数分别为9.55士2.45枚、6.20士0.84枚和6.71士0.61枚,差异不显著(P0.05),但两者与首次超排的差异显著;对供体羊是否完成补饲过程进行对照可知,平均的采胚数不存在显著的差异,不过经过补饲的羊,相对那些没有经过补饲的羊,在平均可用胚数的表达方面,前者远远超过后者,因此认为提高对饲养的监管,可以有助于提高胚胎的成功率;通过LH的作用之后,能够增加排卵数(P0.05),不过平均的采胚数和可用的胚数,无法超过没有作用的组别,同时没有完成受精过程的卵数,也远远多于没有作用的组别,综上所述,LH的刺激无法促进不成熟卵泡的排卵,使得实现受精过程的卵数量减少,从而减少了可用胚数;CIDR法与PG同期发情的效果相比较,虽然平均采胚数没有显著差异(P0.05),却平均可用胚数明显下降(P0.05);应用PG和CIDR法对柯尔克孜羊进行同期发情都有较好的效果(P0.05),其中在高寒牧区使用CIDR+PMSG+PG法对处于非繁殖季节的柯尔克孜羊进行处理后的同期发情率、受胎率及产羔率最好;在相同发育阶段的条件下移植一枚胚胎后,A级胚胎与B级胚胎的移植受胎率之间没有显著差异,B级胚胎与C级胚胎的移植受胎率相比较无显著差异(P0.05),但从受胎率的变化情况看出,随着胚胎质量的下降其移植受胎率也出现明显的下降趋势(P0.05);当选择发情同步差在0h、士12h的供、受体羊进行移植时,其受胎率显著高于士24h移植受胎率,但选择在0h、士12h两个时间段内进行移植时其差异不显著(P0.05)。从上述结果分析可知,在没有足够的,发情同步的受体羊时,为了获得较好的产羔率,选择使用同步差在士12h之内的受体羊是最好。
[Abstract]:Both the sheep superovulation and estrus, or sheep embryo transplantation, although many scholars at home and abroad have been studied for decades, but under the influence of various factors, the results are not ideal. In this study, the Kyrgyz sheep as research object, not only are analyzed in detail. The main factors affecting sheep superovulation and estrus synchronization, and in-depth analysis of the key factors influencing the effects of embryo transfer pregnancy, embryo transplantation technology system was established in alpine pasture sheep. The test results show: like Akeqi County alpine pastoral areas to sheep superovulation and the best time for September in mid October. But in December after superovulation appear less stable trend; the Kirghiz sheep were superovulated with the application of PG+FSH method and CIDR+FSH method, even if the average recovery embryo number does not exist A significant difference, but the average available embryos were calculated by second methods, the number will decrease; the total hormone in the same circumstances, from drug costs, better than the CIDR ten FSH of PG ten FSH application of Kirgiz sheep superovulation; in each group of kerc Zi sheep, through medication role, dosage can have 210IU, 250IU, 270IU, 320IU FSH, realize the processing over rows can be found, the best of each 250IU results, the average recovery of embryos and available embryos are more significant than other groups, (P0.01); first of Kirghiz sheep after superovulation, ovulation between 1 and 2. During the period of superovulation were 9.55 + 2.45, 6.20 + 0.84 and 6.71 + 0.61, the difference was not significant (P0.05), but the difference with the first superovulation significantly; the donor sheep feeding process is completed according to the average number of embryo collection deposit In a significant difference, but after feeding the sheep, relative to those without feeding the sheep, in the expression of the average number of available embryos, the former more than the latter, so that to improve the feeding regulation, can help to improve the success rate of embryo; through the LH function, can increase the number of ovulation (P0.05), but the average recovery of embryos and available embryos, not more than no group, and no number of fertilized eggs of more than not working groups, to sum up, LH stimulation can promote the immature follicle ovulation, which reduces the number of eggs to achieve fertilization process. Reduce the number of available embryos; CIDR and PG compared the effect of estrus, although the average recovery of embryos had no significant difference (P0.05), but the average number of available embryos decreased significantly (P0.05); the Kirgiz sheep estrus using PG and CIDR method Have a good effect (P0.05), estrus in the non breeding season of Kirghiz sheep which use CIDR+PMSG+PG in alpine pastoral area after the rate, pregnancy rate and best lambing rate; at the same developmental stage under the condition of embryo transplantation, there was no significant difference between grade B and grade a embryos embryo transfer pregnancy the rate of grade B and grade C, embryo embryo transfer pregnancy rate compared with no significant difference (P0.05), but the changes of pregnancy rate that decreased with the quality of embryo transfer pregnancy rate also significantly decreased (P0.05); when choosing the estrous synchronization difference in 0h + 12h for receptor the sheep were transplanted, the pregnancy rate was significantly higher than that of 24h pregnancy rate, but in 0h, 12h two time when transplantation no significant differences (P0.05). The analysis results from the above, there is not enough, estrus synchronization receptor Sheep, in order to obtain better lambing rate, choose to use the synchronization error within the 12h receptor in sheep, is the best.
