褪黑素受体在绵羊附睾上的表达

【摘要】:褪黑素(melatonin,Mel)对于动物生殖活动具有重要的调节作用,它通过褪黑素受体MT1、MT2发挥其功能。附睾是雄性动物的一个重要的生殖器官,是精子成熟和贮存的主要场所。本文研究了褪黑素受体在不同年龄绵羊附睾的表达和分布情况,为进一步研究褪黑素对附睾功能的调节提供基础。研究结果如下:(1)通过实时荧光定量PCR检测,MT1在不同年龄绵羊附睾的不同部位均有表达,对于幼龄羔羊(2~3月龄),MT1在附睾尾的转录水平显著高于附睾头和附睾体;青年绵羊(6~8月龄),MT1在附睾体和附睾尾的转录水平显著高于附睾头;成年绵羊(2~3岁),MT1在附睾尾的转录水平显著高于附睾头和附睾体。随着年龄的增加,MT1在附睾部位的转录水平呈现降低的趋势。MT2在不同年龄绵羊附睾的不同部位均有表达,其中幼龄羔羊,附睾头MT2的转录水平显著高于附睾体和附睾尾的;青年绵羊,MT2在附睾头的转录水平极显著高于附睾体和附睾尾,附睾尾的转录水平显著高于附睾体;成年绵羊,MT2在附睾头和附睾尾的转录水平极显著高于附睾体。随着年龄的增加,MT2在附睾部位的转录水平呈现升高的趋势。(2)通过免疫组化技术对不同年龄绵羊附睾的不同部位的褪黑素受体蛋白分布进行分析,结果表明,MT1和MT2蛋白在各个年龄绵羊附睾的各个部位都有分布,且主要分布在附睾上皮细胞中,而且在青年绵羊和成年绵羊的附睾腔以及精子上也检测到MT1和MT2蛋白的阳性信号。总之,在不同年龄绵羊附睾的不同部位都有MT1和MT2的存在,而且表达水平和分布状态存在一定差异。总体上,随着年龄的增加,绵羊附睾上MT1的表达呈下降趋势,而MT2的表达呈升高趋势。这也提示对于不同年龄绵羊,附睾中褪黑素可能通过MT1或/和MT2不同途径发挥生理功能。
[Abstract]:Melatonin (melatonin) plays an important role in regulating animal reproductive activity through melatonin receptor MT1. The epididymis is an important reproductive organ of male animals and the main site for sperm maturation and storage. The expression and distribution of melatonin receptor in epididymis of sheep of different ages were studied. In order to provide a basis for further study on the regulation of melatonin on epididymal function. The results are as follows: 1) the expression of mt1 in different parts of epididymis of sheep of different ages was detected by real-time fluorescence quantitative PCR. The transcriptional level of MT1 in the tail of epididymis was significantly higher than that in the head of epididymis and the body of epididymis. The transcriptional level of MT1 in the epididymis and cauda epididymidis was significantly higher in young sheep than in the head of epididymis. The transcriptional level of MT1 in the caudal epididymis of adult sheep was significantly higher than that of the head of epididymis and the body of epididymis. The transcriptional level of MT1 in epididymis showed a decreasing trend. MT2 was expressed in different parts of epididymis of sheep of different ages, including young lambs. The transcription level of MT2 in the epididymal head was significantly higher than that in the epididymal body and cauda epididymidis. The transcriptional level of MT2 in the epididymal head was significantly higher than that in the epididymal body and cauda epididymidis, and the transcriptional level of the epididymal tail was significantly higher than that of the epididymal body. The transcriptional levels of MT2 in the head and tail of epididymis were significantly higher in adult sheep than in the body of epididymis. The distribution of melatonin receptor protein in different parts of epididymis of sheep of different ages was analyzed by immunohistochemistry. MT1 and MT2 proteins are distributed in all parts of epididymis of sheep of all ages, and are mainly distributed in epididymal epithelial cells. The positive signals of MT1 and MT2 protein were also detected in epididymal cavity and sperm of young sheep and adult sheep. In short, MT1 and MT2 were found in different parts of epididymis of sheep of different ages. On the whole, with the increase of age, the expression of MT1 in sheep epididymis showed a downward trend. However, the expression of MT2 increased, which suggested that melatonin in epididymis may play a physiological role through MT1 or / and MT2 in different age sheep.
