捕食线虫性真菌-少孢节丛孢菌冻干生物制剂的制备

【摘要】:通过在含有0.05%琼脂粉的沙氏葡萄糖肉汤培养基中培养少孢节丛孢菌(Arthrobotrys oligospora)再转接到玉米粒培养基的方法,对少孢节丛孢菌分生孢子进行培养。在不同时间点,将培养好的分生孢子从培养基上洗脱并进行计数,然后制成冻干制剂,并观察其复苏情况,从而为少孢节丛孢菌冻干制剂的临床应用提供基础数据。结果显示:通过对少孢节丛孢菌冻干制剂在萌发率、生长率和捕食率三方面的复苏情况观察,少孢节丛孢菌冻干后的分生孢子在接种后4d,萌发率达到最大值79.5%,平均生长率为3.4mm/d,加入虫体后7d捕食率最高,为95.8%,平均每天捕食率为74.0%;少孢节丛孢菌冻干制剂在平均生长率和捕食率方面与对照组相比,差异均不显著(P0.1)。结果表明,冻干制剂材料易得,方法简单易行,复苏效果良好,进一步优化后将具有产业化应用的前景。
[Abstract]:Arthrobotrys oligospora was cultured in a broth medium containing 0.05% Agar powder. The method of transferring to the corn kernel medium. At different time points, the cultured conidia were eluted from the culture medium and counted, then the lyophilized preparation was prepared and its recovery was observed. The results showed that the recovery rate, growth rate and predation rate of lyophilized preparation were observed in three aspects: germination rate, growth rate and predation rate. The germination rate of the lyophilized conidia reached a maximum of 79.5 mm / d at 4 days after inoculation, and the average growth rate was 3.4 mm / d, and the predation rate was the highest (95.8%) at 7 days after inoculation. The average daily predation rate was 74.0%; Compared with the control group, the average growth rate and predation rate of the lyophilized preparation were not significantly different from those of the control group (P 0.1). The results showed that the freeze-dried preparation was easy to obtain, the method was simple and feasible, and the effect of resuscitation was good. Further optimization will have the prospect of industrial application.
