枯草芽孢杆菌中表达的Cec Md及其衍生抗菌肽的活性研究

【摘要】:为了检测在枯草芽孢杆菌中表达的Cec Md及其衍生抗菌肽的生物活性,研究采用微生物试验技术,对从枯草芽孢杆菌中表达后纯化出的抗菌肽Cec Md及其衍生抗菌肽CC31、CC34的抑菌效果、热稳定性、最佳抗菌活性的pH值、保存稳定性以及蛋白酶敏感性进行了测定。结果表明:CC31、CC34和Cec Md对大肠杆菌、鸡沙门氏菌和停乳链球菌有抑制作用;对有益菌短乳杆菌、德氏乳杆菌、卷曲乳杆菌、双歧杆菌、毕赤酵母SMD1168、毕赤酵母GS115均没有抑制作用;3种抗菌肽对大肠杆菌、鸡沙门氏菌和停乳链球菌的最小抑菌浓度(MIC)均低于25μmol/L,CC31和CC34对金黄色葡萄球菌的MIC为25μmol/L,低于Cec Md(100μmol/L);CC31和CC34对致病菌的最小杀菌浓度(MBC)均不超过50μmol/L,Cec Md对停乳链球菌的MBC为50μmol/L,对其他3种致病菌的MBC均在75μmol/L以上。CC31和CC34的热稳定性较好,Cec Md在沸水中30 min失去抑菌活性;CC31和CC34在pH值低于6时均活性较好,Cec Md最佳活性pH值仅在4~5之间;3种抗菌肽的最佳保存温度均为-80℃;3种抗菌肽经胰蛋白酶、胃蛋白酶、蛋白酶K作用后活性均有所降低,其中蛋白酶K对3种抗菌肽活性影响较弱。枯草芽孢杆菌中表达出的抗菌肽CC31和CC34的抑菌活性与理化特性等方面都好于Cec Md,其中CC31的生物活性最好。
[Abstract]:In order to detect the bioactivity of Cec Md and its derived antimicrobial peptides expressed in Bacillus subtilis, the antimicrobial peptide Cec Md and its derived antimicrobial peptide CC31C34 were purified from Bacillus subtilis by microbiological test technique. The thermal stability, pH value of the best antibacterial activity, preservation stability and protease sensitivity were determined. The results showed that: 1. CC31CC34 and Cec Md could inhibit Escherichia coli, Salmonella chickensi and Streptococcus acuminatum, while they could inhibit the beneficial bacteria Lactobacillus brevis. Lactobacillus de, Lactobacillus coreus, Bifidobacterium, Pichia pastoris SMD1168 and Pichia pastoris GS115 had no inhibitory effect on Escherichia coli. The minimum inhibitory concentrations (MIC) of Salmonella and Streptococcus staphylococcus were lower than 25 渭 mol / L CC31 and CC34, the MIC of Staphylococcus aureus was 25 渭 mol / L, and the minimum bactericidal concentration of Cec Md(100 渭 mol / L CC31 and CC34 were not more than 50 渭 mol / L LCec Md, respectively. The MBC of Streptococcus acuminatum was less than 50 渭 mol / L LCec Md. 50 渭 mol / L, the MBC of the other three pathogens were all above 75 渭 mol/L. CC31 and CC34 had better thermal stability. CecMd lost its antibacterial activity in boiling water for 30 min. CC31 and CC34 had better activity when pH was lower than 6:00. The optimum pH value of CEC Md was only between 4 and 5.
