广西A型流感病毒的耐药性研究

【摘要】:本研究使用氨基酸耐药性位点分析与微量细胞病变抑制法相结合的方法对广西区近年来18株猪流感病毒H1N1和1株马流感病毒H3N8亚型进行耐药性研究。通过对这些毒株进行NA与M基因的克隆测序,基因序列的比对,序列分析结果显示,18株猪流感病毒H1N1亚型的M2基因耐药性位点上均发生S31N突变;1株马H3的毒株耐药性位点没有发生突变。在NA酶活性位点及周围相关位点上,广西区近年来18株猪流感病毒H1N1毒株中,GX3发生了E119K的突变,6号发生E119G突变,G2发生N295D突变,其余的15株H1N1流感病毒均无发生突变;马流感H3N8毒株发生S247A突变。微量细胞病变抑制法实验中,使用浓度为1mg/ml的盐酸金刚烷胺能抑制毒株马H3感染MDCK细胞,该毒株对该浓度下的盐酸金刚烷胺敏感,药物有效浓度为10-1.5 mg/ml;18株猪流感病毒对该浓度的盐酸金刚烷胺耐药,MDCK细胞均被感染,感染后发生病变。微量细胞病变抑制法实验中,使用浓度为1mg/,l的磷酸奥司他韦,能抑制18株猪流感病毒感染MDCK细胞,这些毒株对该浓度下的磷酸奥司他韦敏感,药物有效浓度在10-1~10-2.76mg/ml之间;毒株马H3对该浓度的磷酸奥司他韦耐药,MDCK细胞在被感染后发生细胞病变。
[Abstract]:In this study, amino acid resistance site analysis combined with microcytopathic inhibition was used to study the resistance of 18 swine influenza viruses (H1N1) and 1 equine influenza virus (H3N8) subtypes in Guangxi in recent years. Na and M genes were cloned and sequenced. The results of sequence analysis showed that S31N mutation was not found in the resistant loci of M2 gene of swine influenza virus H1N1 subtype and that of horse H3 strain H3 strain, and no mutation was found at the Na-enzyme activity site and the related loci around the Swine Influenza virus H1N1 subtype, the results of sequence analysis showed that there was no mutation in the resistance loci of S31N mutation. In recent years, the mutation of E119K was found in GX3 of 18 swine influenza virus H1N1 strains in Guangxi, N295D mutation occurred in G _ 2 of G _ 2 mutation of E119G mutation on 6, and no mutation was found in the other 15 strains of H1N1 influenza virus. In the microcytopathic inhibition assay, a dose of 1 mg / ml of amantadine hydrochloride could inhibit the infection of equine H3 to MDCK cells, and the strain was sensitive to amantadine hydrochloride at that concentration. The effective concentration of the drug was 10-1.5 mg / ml ~ (-1) mg / ml ~ (-1) mg 路ml ~ (-1) 路ml ~ (-1) mg 路ml ~ (-1) 路ml ~ (-1) mg 路ml ~ (-1) mg 路ml ~ (-1) mg 路ml ~ (-1) mg 路ml ~ (-1) mg 路ml ~ (-1) mg / L of oseltamivir phosphate. These strains were sensitive to oseltamivir phosphate at that concentration, and the effective concentration of the drug was between 10-1 and 10-2.76 mg / ml. Cytopathic effect of horse H _ 3 strain on oseltamivir phosphate resistant MDCK cells was observed after infection.
