浏阳黑山羊捻转血矛线虫感染情况调查及线粒体部分基因分析

【摘要】:本研究在对浏阳羊捻转血矛线虫感染情况的基础上,对捻转血矛线虫进行了形态学和分子生物鉴定,旨在为防治羊线虫病提供科学依据。本研究的第一部分是采用剖检方法调查浏阳东南西北以及城区的黑山羊捻转血矛线虫感染情况,结果表明东南西北城区感染率分别为:75.00%、66.67%、57.14%、40.00%和35.71%;其中东区黑山羊捻转血矛线虫感染率最高为75.00%,城区采集的黑山羊捻转血矛线虫感染率最低为35.71%。本研究的第二部分对浏阳市黑山羊捻转血矛线虫线粒体cox1, cox3, cytb基因的序列分析,结果发现本研究中的线粒体cox1, cox3, cytb基因片段,coxl基因A+T碱基含量为64.14%-65.97%,G+C碱基为34.03%-35.87%;cox3基因A+T碱基含量为69.58%-70.87%,G+C碱基为29.13%-30.42%;cytb基因的A+T碱基含量为72.30%-74.37%,G+C碱基为25.69%-27.71%。利用Mega 4.1软件对7株捻转血矛线虫进行序列分析,结果显示七株捻转血矛线虫voxl基因序列种内变异率0.9--4.8%,种间9.4-16%;cox3基因序列种内变异率1.2--6.9%;cytb基因序列种内变异率1.8—8.2%。
[Abstract]:In this study, morphological and molecular biological identification were carried out on the basis of the infection of the blood spear nematode in Liuyang sheep. In order to provide scientific basis for the prevention and control of sheep nematode disease, the first part of this study was to investigate the infection of blood nematode twirling in black goats in Liuyang, east, south, northwest and urban areas by using dissecting method. The results showed that the infection rate was 40.00% and 35.71in the east east black goat and the black goat in the east, and the lowest infection rate was 35.71.The second part of this study was about the second part of this study, and the lowest infection rate of the black goat was 35.71.The second part of this study was about the second part of this study. Sequence analysis of mitochondrial cox1, cox3 and cytb genes of Haemonema japonicus in Yangshi Black Goat. The results showed that the content of mitochondrial cox1, cox3, cytb gene A T base was 64.14- 65.97g C base and 34.03-35.87Cox3 gene A T base content was 69.58-70.87G C base was 29.1313-30.42cyt b gene A T base content was 72.30-74.37G C base was 25.-27.71.The content of Mega 4.1 was 25.-27.71. The software was used to analyze the sequence of 7 strains of blood nematode twisting. The results showed that the intraspecific variation rate of voxl gene sequence was 0.9-4.8, and that of 9.4-16and cox3 gene sequence was 1.2--6.9. the variation rate of voxl gene sequence was 1.8-8.2%.
