卫气补对老年鼠免疫、抗氧化功能的影响及其急、慢性毒理试验研究

【摘要】:中药是传统医学的瑰宝。大量研究证实,许多中药具有增强机体免疫力、抗氧化、抗肿瘤的功效,本试验以卫气补(WQB)为研究对象,研究其对老年鼠的免疫及抗氧化功能的影响。通过急、慢性毒性试验,研究WQB对青年鼠的安全性,为WQB的临床应用提供理论依据。研究中药复方卫气补(WQB)对老年鼠免疫指标及抗氧化功能的影响。50只12月龄的老年鼠,随机分为5组,其中A组为对照组,B组饲喂10mg/kg BW左旋咪唑组,C、D、E组分别为0.1%、1%、2%WQB组,另取10只2月龄青年鼠作为F组,正常饲喂。分别按照上述分组饲喂30d。结果显示,与对照组及左旋咪唑组相比,饲料中添加1%WQB能够增加老年鼠体重及免疫器官肝、脾、胸腺指数,显著提高鼠体内SOD以及IL-2水平并降低MDA含量(P0.05,P0.01);增加空肠粘膜固有层CD4+阳性细胞数量、抑制CD8+阳性细胞的表达,增加十二指肠s Ig A细胞的表达量;饲料中添加1%、2%WQB均能显著提高老年鼠血清中IFN-γ含量,并达到青年鼠水平。为评价WQB安全性,进行WQB急性毒性试验研究。将80只2月龄青年鼠随机分为7个剂量组分别为2 m L/kg BW、2.80 m L/kg BW、3.92 m L/kg BW、5.49 m L/kg BW、7.68 m L/kg BW、10.76 m L/kg BW、15.06 m L/kg BW及1个灌服生理盐水的空白对照组,灌服1周后,各组小鼠均表现正常,没有死亡情况,因此又进行最大耐受剂量试验(MTD),灌服剂量为30.12 m L/kg BW,相当于每只小鼠灌服0.9g原药。饲喂1周后,小鼠表现正常,无死亡情况。取上述9组小鼠肝、肾器官称重;由于在急性毒性试验期间,WQB各组小鼠没有死亡,所以摘取15.06 m L/kg BW WQB组、空白对照组及MTD组小鼠肝、脾、肺、肾、十二指肠等器官,观察其组织结构;眼眶采血,测定血常规。结果表明,各组小鼠肝、肾系数之间在0.05和0.01水平上无统计学意义;15.06 m L/kg BW WQB组及MTD组小鼠脏器组织结构与空白对照组小鼠相比,结构正常;血常规指标如MID(中间细胞)、LYM(淋巴细胞)、RBC(红细胞)、HGB(血红蛋白)检测结果,各组均无显著性差异(P0.05)。为评价WQB安全性,进一步进行WQB慢性毒性试验研究。将15只2月龄青年鼠随机分为3组,分别为空白对照组:正常饲喂;阳性对照组:饲喂10 mg/kg BW苯丙左旋咪唑;1%WQB组:在饲料中加拌1%WQB。饲喂90d后取材,摘取肝、肾组织并称重;由于在慢性毒性试验期间,小鼠没有死亡,所以选取空白对照组、1%WQB组小鼠肝、脾、肺、肾、十二指肠组织固定,用于观察组织结构。眼眶采血用于血生化指标甘油三酯(TG)、胆固醇(CHOL)、葡萄糖(GLU)、尿酸(UA)、肌酐(Crea)、丙氨酸转氨酶(ALT)、尿素氮(BUN)、总蛋白(TP)、白蛋白(ALB)、天冬氨酸转氨酶(AST)的检测。结果表明,各组小鼠生长良好,灌服1%WQB组小鼠免疫器官指数及体重变化与正常对照组及苯丙左旋咪唑组相比,无显著性差异;1%WQB组小鼠与正常对照组小鼠脏器组织结构相比,无变化;1%WQB组小鼠血生化指标与空白对照组和苯丙左旋咪唑组小鼠相比,差异不显著(P0.05)。本试验表明饲料中加入1%WQB可提高老年鼠免疫力及抗氧化能力,且安全无毒。
[Abstract]:Chinese medicine is a treasure of traditional medicine. A number of studies have demonstrated that many Chinese medicine has to enhance immunity, anti-oxidation, anti-tumor effect, the Wei Qi supplement (WQB) as the research object, to study the influence of old mice's immune function and antioxidation. The acute, chronic toxicity test, safety study of WQB in young rats, and provide a theoretical basis for the clinical application of WQB. The study of compound traditional Chinese medicine Qi supplement (WQB) effect of.50 on immune function and antioxidant function in aged rats only 12 month old aged rats, were randomly divided into 5 groups, group A as control group, B group fed with 10mg/kg BW levomisole group, C. D, E group were 0.1%, 1%, 2%WQB group, another 10 male 2 month old young rats as F group, normal feeding. The feeding group respectively according to the 30D. results showed that compared with the control group and levamisole group, 1%WQB supplementation can increase the weight of immune organs and liver spleen in aged rats, Thymus index increased significantly in rats SOD and the level of IL-2 and decrease the content of MDA (P0.05, P0.01); increase the number of CD4+ positive cells in jejunum mucosa, inhibit the expression of CD8+ positive cells, increase the expression of s Ig A cells in the duodenum; add 1% feed, 2%WQB could significantly increase the IFN- content of serum gamma in aged rats in young rats and reached the level. In order to evaluate the safety of WQB, experimental study on acute toxicity of WQB. 80 2 month old young rats were randomly divided into 7 groups respectively for 2 m L/kg BW m L/kg, 2.80 BW, 3.92 m L/kg BW m L/kg, 5.49 BW, 7.68 m L/kg BW 10.76, m L/kg BW m L/kg, 15.06 BW and 1 normal saline control group, gavage after 1 weeks, the mice showed normal, no death case, and thus the maximum tolerated dose test (MTD), dose 30.12 m L/kg BW, the equivalent of every mice fed with 0.9g TC. The results showed that the mice liver, kidney coefficient between 0.05 and 0.01 levels was not statistically significant; 15.06 m L/kg BW WQB group and MTD group of mice organ structure and the blank control group were compared with normal structure; blood routine indexes such as MID (intermediate cell) LYM, RBC (lymphocytes), (RBC), HGB (hemoglobin) test results, there were no significant differences between the groups (P0.05). To evaluate the safety of WQB, further experimental study of WQB chronic toxicity. 15 2 month old young rats were randomly divided into 3 groups, namely control group: Normal feeding; positive control group: rats were fed with 10 mg/kg BW styrene acrylate levamisole; group 1%WQB: 1%WQB. add mix feeding 90d in feed harvested after removal of the liver, kidney and weighed; because during the chronic toxicity test, mice without death, so select the blank control group, 1%WQB group of mice liver, spleen and lung. Kidney, duodenal tissue fixed for histological observation. Blood biochemical index for triglyceride (TG), cholesterol (CHOL), glucose (GLU), uric acid (UA), creatinine (Crea), alanine aminotransferase (ALT), urea nitrogen (BUN), total protein (TP), albumin (ALB), aspartate aminotransferase (AST) detection. The results showed that the mice fed with good growth, immune organ index and body weight in 1%WQB group compared with the normal control group and phenylpropanoid levamisole group, no significant difference; in 1%WQB group compared with the normal control group mice. No change; 1 Compared with blank control group and phenylpropanolone group, there was no significant difference in blood biochemical index between%WQB group and P0.05 group (P0.05).
