牛磺鹅去氧胆酸对大鼠脂肪代谢的影响

【摘要】:胆汁酸(blie acid)是动物体内胆汁中的主要成分,直接参与体内脂类代谢过程。牛磺鹅去氧胆酸(Taurochenodeoxycholic Acid,TCDCA)是胆汁酸中主要的生物活性物质,主要存在于鸡胆汁中,具有抗氧化、解热、抗炎、免疫、镇痛等多种生物功能。为了深入了解TCDCA的生物活性,本课题针对TCDCA对脂肪代谢的影响进行了试验研究。研究内容包括:经口服给予不同剂量的TCDCA 21d后,采用全自动生化分析仪检测TCDCA对大鼠脂肪代谢的相关生理生化指标;分别采用TBA法、WSF-1法和分光光度法测定血清中丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活力;采用组织病理学方法检测大鼠肝脏组织形态和细胞结构;采用免疫组化方法检测法尼醇X受体(FXR)在大鼠肝脏的表达位置;分别采用荧光定量PCR和Western blot技术检测大鼠肝脏中FXR基因、蛋白的表达量。研究结果表明:中剂量组的大鼠体重及增重均显著低于阴性对照组(P<0.05);低、中、高三个剂量组肝体比极显著大于阴性对照组(P<0.01);与阴性对照组相比,TCDCA低、中、高三个剂量组的TC、TG、HDL-C,LDL-C水平、肾体比、体脂肪率、料肉比以及各项血液生理生化指标均无显著性差异(P>0.05);中剂量TCDCA可显著性降低大鼠血清中MDA含量,TCDCA处理组的大鼠血清中S0D和GSH-Px活力虽有升高,但差异不显著(P>0.05);TCDCA给药组的大鼠肝脏与其正常组织形态结构相比均无显著变化(P>0.05);免疫组化结果显示,FXR在大鼠肝脏细胞核上均有表达,但TCDCA给药组的大鼠肝脏中FXR的表达量较多;TCDCA低剂量组的大鼠肝脏中FXR mRNA表达量极显著高于阴性对照组(P<0.01),Western blot结果显示TCDCA低剂量组FXR蛋白表达量显著高于阴性对照组(P<0.05)。结论:中剂量TCDCA显著降低大鼠体质量,显著抑制大鼠增重;FXR在大鼠肝脏中有较高效表达,且TCDCA处理组的大鼠肝脏中FXR表达量明显高于阴性对照组:低剂量TCDCA能够显著性升高FXR的蛋白和基因的表达量。
[Abstract]:Taurochenodeoxycholic acid (Taurochenodeoxycholic acid TCDCAA) is the main bioactive substance in bile acid, which mainly exists in chicken bile. In order to understand the biological activity of TCDCA, the effects of TCDCA on fat metabolism were studied. The content of malondialdehyde (MDA) in serum was determined by TBA method and spectrophotometry, respectively, using automatic biochemical analyzer to detect the physiological and biochemical indexes related to fat metabolism of rats by TCDCA, and to determine the content of malondialdehyde (MDA) in serum by TBA method and spectrophotometry, respectively. The activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were detected by histopathological method, and the expression of FXR in rat liver was detected by immunohistochemical method. Fluorescence quantitative PCR and Western blot were used to detect the expression of FXR gene and protein in the liver of rats. The results showed that the weight and weight gain of middle dose group were significantly lower than those of negative control group (P < 0.05, P < 0.05, P < 0.05, P < 0.05). The liver body ratio of the three dose groups was significantly higher than that of the negative control group (P < 0.01), and the TCDCA was lower than that of the negative control group. There was no significant difference in feed / meat ratio and blood physiological and biochemical indexes (P > 0.05), and moderate dose TCDCA could significantly decrease the content of MDA in serum and increase the activity of S0D and GSH-Px in serum of rats treated with TCDCA-treated group. However, there was no significant difference between the two groups (P > 0.05) and the expression of FXR was found in the nuclei of the liver of the rats treated with TCDCA (P > 0.05), but there was no significant difference between the two groups (P > 0.05). However, the expression of FXR mRNA in the liver of the TCDCA group was significantly higher than that of the negative control group (P < 0.01). The results showed that the expression of FXR protein in the low-dose TCDCA group was significantly higher than that in the negative control group. Conclusion: middle dose TCDCA can significantly reduce the body mass of rats. The expression of FXR in the liver of rats treated with TCDCA was significantly higher than that in the control group. Low dose TCDCA could significantly increase the expression of FXR protein and gene.
