抗炎中药“宫炎净”复方的优化研究

【摘要】:目的:通过中兽医实验室前期研究发现,中药复方“宫炎净”主要有效成分为生物碱,本试验拟通过比较中药复方“宫炎净”、简化复方、君药益母草中的生物碱对奶牛子宫内膜炎主要致病菌金黄色葡萄球菌、大肠杆菌及无乳链球菌的抑菌效果,以及对小鼠耳廓肿胀、足趾肿胀、腹腔毛细血管通透性以及肉芽肿炎症模型的抗炎效果,为优化中药复方“宫炎净”提供理论依据。方法:1.将粗提的自拟中药复方“宫炎净”各单味药(益母草、川芎、当归、桃仁、甘草、五灵脂、干姜、蒲黄)中加入碘化汞钾试剂、碘化铋钾试剂、硅钨酸试剂、磷钼酸试剂,检测每味药中是否含有生物碱,对初步筛选含有生物碱的各单味药进行精制,将含有生物碱的单味药按原方剂量配制为简化复方。2.通过乙醇回流提取法与超声波辅助提取法分别提取中药复方“宫炎净”、简化复方、君药益母草中的生物碱。3.通过平板打孔法和试管二倍稀释法测定中药复方“宫炎净”、简化复方、君药益母草中的生物碱对奶牛子宫内膜炎主要致病菌金黄色葡萄球菌、无乳链球菌、大肠杆菌体外抑菌效果。4.通过建立小鼠耳廓肿胀、足趾肿胀、腹腔毛细血管通透性以及肉芽肿炎症模型来评估中药复方“宫炎净”、简化复方、君药益母草中生物碱的抗炎效果。结果:1.自拟中药复方“宫炎净”8味药粗提液分别加入生物碱试剂检测发现,益母草、川芎、甘草中可能含有生物碱,通过进一步精制后确定上述三味药含有生物碱,将益母草、川芎、甘草按照原方剂量配制为简化复方。2.通过对比乙醇回流提取法与超声辅助提取法发现,自拟中药复方“宫炎净”、简化复方、君药益母草中的生物碱,乙醇回流提取法提取率分别为0.92%、0.93%、0.71%,超声辅助提取法提取率分别为0.87%、0.86%、0.62%。3.通过体外抑菌试验,发现自拟中药复方“宫炎净”、简化复方、益母草中的生物碱对金黄色葡萄球菌、大肠杆菌及无乳链球菌的平均抑菌圈直径分别为17.4±0.8mm、16.9±1.2mm、7.8±0.8mm,上述三组生物碱对金黄色葡萄球菌MIC、MBC分别为32 g/L、125 g/L、500g/L,62.5 g/L、125 g/L、500g/L;对大肠杆菌MIC、MBC分别为62.5 g/L、125 g/L、250g/L,125 g/L、250 g/L、500g/L;对无乳链球菌MIC、MBC分别为125 g/L、125 g/L、500g/L,250 g/L、250g/L、未测出。4.对小鼠的抗炎试验发现中药复方“宫炎净”、简化复方及益母草中的生物碱均对小鼠耳廓肿胀、足部肿胀、腹腔毛细血管通透性和棉球肉芽肿的形成有抑制作用,其中复方“宫炎净”和简化复方的抗炎效果与益母草生物碱组差异显著(P0.05),与地塞米松组差异不显著(P0.05),简化复方的抗炎效果略优于复方“宫炎净”。结论:简化复方与自拟中药复方“宫炎净”的体外抑菌作用、体内抗炎活性均相差不大,为简化复方的临床运用提供了理论依据。
[Abstract]:Objective: to find out that the main active component of the compound "Gong Yan Jing" is alkaloid through the preliminary study of Chinese Veterinary Laboratory. This experiment aims to simplify the compound by comparing the traditional Chinese medicine compound "Gong Yan Jing". The inhibitory effect of alkaloids in Herba Leonurus on Staphylococcus aureus, Escherichia coli and Streptococcus acuminatum, the main pathogens of endometritis in dairy cows, as well as the swelling of auricle and toes in mice, The permeability of celiac capillaries and the anti-inflammatory effect of granulomatous inflammatory model provide the theoretical basis for optimizing the traditional Chinese medicine compound "Gong Yan Jing". Methods: 1. The crude extracted Chinese medicine compound "Gong Yan Jing" (Leonurus chinensis, Ligusticum chuanxiong, Angelica sinensis) was prepared. Peach kernel, licorice, WuLingzhi, dried ginger, fuhuang) were added to each drug to determine whether there were alkaloids in each drug, such as potassium mercuric iodide, potassium bismuth iodide, silicotungstic acid and phosphomolybdic acid. The primary screening of each single drug containing alkaloids was carried out, The single medicine containing alkaloids was prepared as simplified compound compound according to the original dosage. The compound was extracted by ethanol reflux extraction method and ultrasonic assisted extraction method respectively. Alkaloids in Herba Leonurus mongolicum. The alkaloids in Herba Leonurus chinensis were determined by plate perforation method and double dilution method of test tube to simplify and simplify the alkaloids of Herba Leonurus to staphylococcus aureus, the main pathogen of endometritis of dairy cow. No Streptococcus lactobacillus, Escherichia coli in vitro bacteriostatic effect .4. by establishing mouse models of auricle swelling, toe swelling, celiac capillary permeability and granulomatous inflammation to evaluate the compound "Gong Yan Jing" and simplify the compound, The antiinflammatory effect of alkaloids in Herba Leonurus chinensis. Results 1. The crude extracts of 8 kinds of Chinese medicine compound "Gong Yan Jing" were tested with alkaloid reagent respectively. It was found that the alkaloids may exist in Leonurus, Ligusticum chuanxiong and Glycyrrhiza uralensis. After further refining, the above three drugs were confirmed to contain alkaloids, and the three drugs were made up of Leonurus chinensis, Ligusticum chuanxiong and Glycyrrhiza uralensis according to the dosage of the original prescription to simplify compound compound .2.The results were found by comparing the ethanol reflux extraction method with the ultrasonic assisted extraction method. The extraction rates of alkaloids in Herba Leonurus and ethanol reflux extraction were 0.92% and 0.73%, respectively. The extraction rate of ultrasonic assisted extraction method was 0.87%, 0.86%, 0.62% and 0.32%, respectively. The in vitro bacteriostasis test was carried out. It was found that the traditional Chinese medicine compound "Gong Yan Jing", simplified compound, Leonurus alkaloids in Staphylococcus aureus, The average bacteriostasis diameters of Escherichia coli and Streptococcus lactobacillus were 17.4 卤0.8mm, 16.9 卤1.2mm and 7.8 卤0.8mm, respectively. The anti-inflammatory effect of compound "Gong Yan Jing" and simplified compound is significantly different from that of Leonurus alkaloid group (P 0.05), but not to dexamethasone group (P 0.05). The anti-inflammatory effect of simplified compound is slightly better than that of compound "Gong Yan Jing". Conclusion: simplified compound is better than that of compound "Gong Yan Jing". And the bacteriostatic effect of "Gong Yan Jing", a traditional Chinese medicine compound, in vitro, The anti-inflammatory activity in vivo is not different, which provides a theoretical basis for simplifying the clinical application of compound.
