不同遗传基础公鹅繁殖性能差异比较及分子水平验证

【摘要】:本试验结合养鹅生产中存在的技术难题,设计了5个不同遗传基础的公鹅试验组。通过饲养孵化试验,检测了试验组间的受精率和使用时间上的差异;通过HE染色组织切片法,比较了繁殖期内不同试验组公鹅睾丸组织形态学变化的差异;通过基因差异表达分析,比较了不同试验组GnRH和PRL的mRNA差异表达。从而为公鹅的繁殖性能优化选择提供了理论参考依据。研究结果如下:(1)组间公鹅受精率:繁殖前期差异显著(p㩳0.05),以Ⅴ组公鹅最高,繁殖中期差异不显著(p㧐0.05),平均值以Ⅴ组公鹅最大。繁殖末期差异显著(p㩳0.05),以Ⅴ组公鹅受精率下降最慢;有效配种天数:Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ组公鹅分别为99、81、99、81、118d,Ⅴ组公鹅较Ⅰ、Ⅱ、Ⅲ、Ⅳ组公鹅有效配种天数分别延长19、37、19、37d。(2)繁殖期内公鹅睾丸组织形态学变化:繁殖初期5个试验组公鹅的睾丸重、曲细精管直径、生殖上皮高度、精子密度等指标均呈现上升现象;繁殖中期,组间公鹅睾丸的各指标增加值虽不相同,但均在此时期达到了最大值;繁殖末期,Ⅰ、Ⅱ、Ⅲ、Ⅳ组公鹅睾丸指标值下降较快,Ⅴ组公鹅睾丸指标值下降相对较慢,与此时期受精率一致。(3)GnRH和PRL表达变化:整个繁殖期内,两基因在5个试验组公鹅睾丸内均有表达,且规律相同。从繁殖初期、中期至末期,GnRH表达量呈现出先增多后减少,繁殖中期表达最高;PRL表达量为先减少后增多,繁殖中期表达最低。GnRH对公鹅繁殖性能有促进作用,PRL则起抑制作用。
[Abstract]:Combining with the technical problems existing in the production of geese, five test groups of male geese with different genetic basis were designed.The differences of fertilization rate and using time among the test groups were detected by feeding and hatching experiment, and the morphological changes of testis in different test groups were compared by HE staining tissue section method.MRNA differential expression of GnRH and PRL in different groups was compared by gene differential expression analysis.Thus it provides a theoretical reference for the selection of reproductive performance of male geese.The results were as follows: fertilization rate of male geese in the early stage of reproduction was significantly different from that in group V, the highest in group 鈪,
