ChREBP在马身猪和大白猪三种组织中的差异性表达研究

【摘要】:ChREBP作为一种转录因子,主要结合在糖酵解和脂肪合成相关基因的启动子上,调控糖脂代谢相关酶的转录。本研究利用qRT-PCR和Western blotting技术,选取马身猪和大白猪初生至180日龄(1,30,60,90,120,150,180)7个生长点,检测ChREBP mRNA和蛋白在肝脏、背最长肌和背部皮下脂肪等3种组织中的发育性表达规律。结果如下:1. ChREBP mRNA在肝脏和背最长肌中的表达模式均为波浪形。马身猪和大白猪肝脏中表达最高峰分别为30和60日龄;两猪种背最长肌中1日龄的表达量极显著低于其他日龄,分别在60和90日龄表达最高;皮下脂肪组织中,两猪种均高表达于生长后期。品种间比较,该基因的表达量在马身猪肝脏30、90、150、180日龄和背最长肌整个阶段均极显著高于大白猪,肝脏其余阶段和皮下脂肪整个阶段,大白猪显著高于马身猪。组织间比较,马身猪整体表现为:肝脏背最长肌脂肪;大白猪中背最长肌中表达量最少,优势表达组织分两个阶段,生长前期为肝脏,后期为背部皮下脂肪。2. ChREBP蛋白在马身猪肝脏90日龄、大白猪60日龄表达最高;两猪种背最长肌中1日龄的表达量最低,随后急剧增长;皮下脂肪组织中30日龄的表达量最高。品种间比较,该蛋白表达量在马身猪肝脏和皮下脂肪中各阶段均低于大白猪;背最长肌中马身猪的表达量高于大白猪,90日龄表达量除外。组织间比较,两猪种背最长肌中的表达量极显著高于另两个组织,大白猪1日龄和60日龄阶段除外。本文结果表明:ChREBP mRNA和蛋白在不同品种、不同组织和不同日龄中表达具有显著差异,提示该基因对猪体内脂肪沉积的调控起重要作用。
[Abstract]:As a transcription factor, ChREBP mainly binds to the promoter of glycolytic and fat biosynthesis related genes to regulate the transcription of glycolipid metabolism-related enzymes.In this study, qRT-PCR and Western blotting techniques were used to detect the developmental expression of ChREBP mRNA and protein in liver, longissimus dorsi muscle and subcutaneous fat of three kinds of tissues.The result is as follows: 1.The expression pattern of ChREBP mRNA in liver and longissimus dorsi muscle was wavy.The highest expression peaks were 30 and 60 days old in liver of Mashen pig and large white pig, respectively, the expression of 1 day old in the longissimus dorsi muscle of both breeds was significantly lower than that of other days old, and the highest expression was in 60 and 90 days old, respectively; in subcutaneous adipose tissue, the highest expression was found in the subcutaneous adipose tissue.Both breeds were highly expressed in the late stage of growth.The expression level of the gene was significantly higher in the 30 ~ 90150 ~ 180 days old of liver and the whole stage of the longissimus dorsi muscle than in the whole stage of liver and subcutaneous fat, and the expression of the gene was significantly higher in the whole stage of liver and subcutaneous fat than that of the horse body pig.Tissue comparison showed that the overall performance of Ma-Shen pig was: liver longissimus dorsi muscle fat; in large white pig, the expression of longissimus dorsi muscle was the least, and the dominant expression tissue was divided into two stages: liver in pregrowth stage and subcutaneous fat in back in later stage.The expression of ChREBP protein was the highest in the liver of Ma-Shen pig at 90 days old and the highest in the white pig at 60 days old, the lowest in the longissimus dorsi muscle of the two breeds at 1 day old, and the highest in the subcutaneous adipose tissue at the age of 30 days.The expression of this protein was lower in liver and subcutaneous fat than that in white pig, except that in longissimus dorsi muscle was higher than that in white pig at 90 days old.The expression level of the longissimus dorsi muscle was significantly higher in the two pigs than in the other two tissues, except for the 1-day and 60-day-old stage.The results showed that there were significant differences in the expression of ChREBP mRNA and protein in different breeds, tissues and days of age, suggesting that the gene plays an important role in the regulation of fat deposition in pigs.
