建立柱前衍生化-HPLC法测定发酵液中L-蛋氨酸

【摘要】:拟建立一种简单、快速、高效的柱前衍生化HPLC法测定发酵液中蛋氨酸含量。以2,4-二硝基氯苯(CDNB)为衍生化试剂,采用高效液相色谱法,定量测定发酵液中的蛋氨酸含量。色谱条件:流动相0.01 mol/L p H 5.2 Na AC-乙腈(70∶30);检测波长:350 nm;柱温:25℃;流速:0.8 m L/min。结果表明:蛋氨酸含量在10~80μg/m L范围内峰面积与浓度呈现良好的线性关系,R2=0.999 24;检测限和定量限分别为0.1和0.5μg/m;加样回收率为98.7%,并且具有良好的重复性。
