ERK信号在玉米赤霉烯酮诱导的睾丸支持细胞自噬中作用的研究

【摘要】:为探讨玉米赤霉烯酮(ZEA)雄性生殖毒性作用机理,本试验以睾丸支持细胞TM4细胞系为对象,研究了ZEA对睾丸支持细胞自噬及相关信号通路的影响。试验分别以浓度为0、0.1、1、10、20、30 mol/L的ZEA进行染毒,24 h后,利用CCK-8、激光共聚焦,透射电镜、Western-blot等技术检测了ZEA对TM4细胞的存活率、自噬,MAPK信号通路的影响以及MAPK信号通路与自噬的相互关系。免疫荧光结果显示,随着ZEA浓度的升高,TM4细胞内LC3聚点数量显著升高,酸性囊泡的个数增多;Western-blot结果显示,自噬相关蛋白LC3Ⅱ、Beclin-1、ATG5、ATG7表达显著升高,而P62蛋白表达显著下降(P0.05或P0.01);MAPK信号通路中ERK1/2磷酸化水平显著升高(P0.05或P0.01),加入ERK磷酸化抑制剂U0126后,ZEA诱导TM4自噬水平显著下降(P0.05或P0.01)。结果表明,在一定浓度范围内,玉米赤霉烯酮可以诱导TM4细胞产生自噬,且发生自噬与ERK信号通路激活有关。
[Abstract]:In order to investigate the mechanism of male reproductive toxicity of Zea, the effects of ZEA on testicular Sertoli cell (TM4) cell line were studied. The survival rate of TM4 cells, the effect of autophagy MAPK signaling pathway and the relationship between MAPK signal pathway and autophagy were detected by CCK-8, laser confocal and transmission electron microscopy Western-blot after exposure to ZEA at the concentration of 0 ~ 0.1 ~ 1 ~ 10 ~ 20 ~ 30 mol/L for 24 h respectively. The results of immunofluorescence showed that with the increase of ZEA concentration, the number of LC3 aggregates in TM4 cells increased significantly, and the number of acidic vesicles increased.
