三七皂苷S-6的溶血性及体外免疫活性研究

【摘要】:研究三七皂苷S-6的溶血性及体外免疫活性。常规方法提取PNS,大孔树脂柱层析分离纯化得S-6。溶血试验测定红细胞溶血率。脾淋巴细胞毒性试验确定S-6的安全剂量范围,体外脾淋巴细胞增殖反应和IL-2的诱生及含量测定检测S-6的体外免疫活性。S-6溶血性较小,2 mg/mL浓度以下无溶血性;S-6(0.1μg/mL、1μg/mL)显著促进了ConA诱导的小鼠脾淋巴细胞增殖反应,且与PNS和APS组相比,差异不显著;S-6(1μg/mL)极显著地促进了LPS诱导的小鼠脾淋巴细胞增殖反应,且与PNS相比,S-6(1μg/mL)差异显著,其余各个剂量与PNS和APS组相比,差异不显著;S-6(12.5μg/mL、25μg/mL、50μg/mL)显著或极显著地提高了IL-2的含量,且与PNS和APS组相比,差异不显著。S-6安全性好,体外具有较好的细胞免疫活性和诱生细胞因子的能力。
[Abstract]:To study the hemolysis and in vitro immune activity of Panax notoginsenoside S-6. PNSs were extracted by conventional method and purified by macroporous resin column chromatography. The hemolysis rate of red blood cells was measured by hemolysis test. The safety dose range of S-6 was determined by splenic lymphocyte toxicity test. Proliferation of splenic lymphocytes in vitro and induction and determination of IL-2 in vitro the in vitro immunological activity of S-6. S-6 hemolytic activity. S-6 hemolytic activity was less than 2 mg/mL. No hemolytic S-6N 0.1 渭 g / mL 1 渭 g 路mL ~ (-1) significantly promoted the proliferation of spleen lymphocytes induced by ConA in mice. Compared with the PNS and APS groups, the difference was not significant (P < 0.05). The proliferation of splenic lymphocytes induced by LPS was significantly promoted by S-6N (1 渭 g / mL), and the difference was significant compared with that of PNS (P < 0.05). The other doses were compared with those of PNS and APS groups. There was no significant difference in S-6, 12.5 渭 g / mL, 25 渭 g 路mL ~ (-1) or 50 渭 g / mL). Compared with PNS and APS, the difference was not significant. S-6 was safe, and had better cellular immune activity and the ability to induce cytokines in vitro.
