猪气喘病的诊断及其阴性猪群的建立

【摘要】:猪气喘病是由猪肺炎支原体(Mycoplasmahyopneumoniae,Mhp)感染引起的一种慢性呼吸道传染病,以咳嗽、喘气和生长发育迟缓为主要临床特征。该病极难防控,给养猪业带来巨大的损失。如何经济有效的控制猪气喘病一直是困扰全球养猪业的难题,目前研究表明控制该病的最好方法是从源头抓起彻底净化气喘病病原。本研究在结合我们研究室对猪肺炎支原体防控及致病机理研究成果的基础上,借鉴前人的研究经验,通过气喘病阳性猪群的诊断,以气喘病阳性母猪为基础,采用集成联合用药、隔离早期断奶(SEW)和"三点式"生产体系的培育方法,成功建立了 190头气喘病阴性群猪。主要研究内容如下:1、猪气喘病阳性猪群的诊断:选择疑似感染猪肺炎支原体且正在发病的某养殖场,通过流行病学调查、临床症状观察、咳嗽指数统计分析、剖解评分、病原分离培养鉴定、血清抗体监测及鼻拭子中抗原荧光定量PCR方法检测等实验室诊断结果综合分析,确诊该场感染了猪肺炎支原体并发病,为后期气喘病的净化方法及监控诊断得以科学、有效的实施奠定基础。2、气喘病阴性群猪的建立:本研究以上述猪群为基础,先通过妊娠母猪的筛选,母猪程序性用药,再通过SEW技术、屏障隔离系统、独立饲养管理体系及仔猪程序性用药的培育方法,培育获得5批共190头猪,通过猪肺炎支原体血清抗体检测和荧光定量PCR检测鼻拭子中抗原检测,持续4个月的检测,证明新培育的5批猪在35日龄后血清抗体均为阴性,鼻拭子中抗原检测全为阴性,表明集成联合运药、SEW和"三点式"生产管理体系技术可以有效净化猪肺炎支原体,为国内猪气喘病的控制和净化提供理论基础及临床实践经验。
[Abstract]:Swine asthma is a chronic respiratory infectious disease caused by Mycoplasma pneumoniae pneumoniae (Mhp) infection. Cough, wheezing and growth retardation are the main clinical features. The disease is extremely difficult to prevent and control, which brings huge losses to the pig industry. How to control asthma in pigs economically and effectively has always been a difficult problem in pig industry all over the world. Current studies show that the best way to control the disease is to thoroughly purify the pathogen of asthma from the source. Based on the research results of prevention and control of mycoplasma pneumoniae and pathogenesis of mycoplasma pneumoniae in our laboratory, and based on the previous research experience, the diagnosis of positive pigs with asthma disease was carried out, and the results were based on the positive sows with asthma disease. 190 pigs with negative asthma disease were successfully established by using the method of integrated drug use, isolating SEW (early weaning) and "three-point" production system. The main contents of the study are as follows: 1, diagnosis of swine asthma positive pigs: select a breeding farm that is suspected to be infected with Mycoplasma pneumoniae and is developing. Through epidemiological investigation, clinical symptom observation, cough index statistical analysis, dissection score, Comprehensive analysis of laboratory diagnostic results such as pathogen isolation and culture identification, serum antibody monitoring and antigen fluorescence quantitative PCR detection in nasal swab showed that mycoplasma pneumoniae was infected in this field. For the later stage of asthma disease purification methods and monitoring diagnosis can be scientific, effective implementation of the foundation of the establishment of asthma negative pigs: this study is based on the above pig herd, first through the screening of pregnant sows, sows program medication, Then through SEW technology, barrier isolation system, independent feeding management system and breeding method of Piglet programmed medication, 5 batches of 190 pigs were obtained. The detection of antibody to mycoplasma pneumoniae in serum and detection of antigen in nasal swab by fluorescence quantitative PCR for 4 months proved that all the 5 groups of pigs were negative in serum and negative in antigen of nasal swab after 35 days of age. The results show that the integrated technology of SEW and "three-point" production management system can effectively purify mycoplasma pneumoniae and provide theoretical basis and clinical experience for the control and purification of swine asthma disease in China.
