用Ala-Gln二肽替换Gln可提高猪体细胞克隆胚胎发育能力

【摘要】:猪(Sus scrofa)的卵母细胞体外成熟培养以及胚胎体外培养是猪体外受精、体细胞克隆以及胚胎干细胞分离的重要基础。优化卵母细胞以及胚胎的体外培养条件,是提高猪卵母细胞体外成熟效率和胚胎体外发育能力的重要方面。本研究比较了分别添加丙氨酰-谷氨酰胺(alanyl-glutamine,Ala-Gln)和谷氨酰胺(glutamine,Gln)的卵母细胞成熟培养液、胚胎培养液(porcine zygote medium-3,PZM-3)对猪卵母细胞成熟、孤雌激活胚胎和体细胞克隆胚胎发育的影响。实验结果表明,丙氨酰-谷氨酰胺对猪卵母细胞第一极体可见率无明显提升作用,但可以提高猪孤雌胚胎的囊胚率和囊胚细胞数,同时能显著提高猪体细胞克隆胚胎的分裂率和囊胚率。本研究为优化胚胎体外培养条件、提高猪体细胞克隆效率提供了一种新的方法。
[Abstract]:In vitro maturation and in vitro embryo culture of Sus scrofa are the important bases for in vitro fertilization, somatic cloning and isolation of embryonic stem cells. To optimize the culture conditions of oocytes and embryos in vitro is an important aspect to improve the efficiency of in vitro maturation of porcine oocytes and the ability of embryo development in vitro. The effects of alanyl-glutamine (Ala-Gln) and glutamine (glutamine) on the development of porcine zygote medium-3PZM-3 and porcine zygote medium-3PZM-3 on porcine oocytes maturation, parthenogenetic embryos and somatic cloned embryos were compared. The results showed that alanyl-glutamine had no effect on the first polar body visibility of porcine oocytes, but could increase the blastocyst rate and the number of blastocysts in parthenogenetic embryos. At the same time, the cleavage rate and blastocyst rate of porcine somatic cloned embryos were significantly increased. This study provides a new method for optimizing the conditions of embryo culture in vitro and improving the cloning efficiency of porcine somatic cells.
