犬乳腺肿瘤诊断及相关因子检测

【摘要】:乳腺肿瘤是犬常见的肿瘤之一,其高病发率和高死亡率严重危害着犬的身体健康,尽管近年来对犬乳腺肿瘤的防治方案不断改进完善,但治愈率并没有得到明显提高,究其原因主要是由于该病临床就诊时多已是中晚期,因此能否提高早期诊断对该病治疗有着重要意义。犬作为人类伴侣动物,其乳腺肿瘤的发生对于研究人类乳腺肿瘤具有一定的参考价值,本实验以犬乳腺肿瘤为研究对象,开展了犬乳腺肿瘤诊断及相关预测因子检测分析的研究。1.犬乳腺肿瘤临床病例调查分析:调查患犬病史资料、观察临床症状和详细记录诊疗的过程,统计分析发病犬的品种、性别、年龄、生育状况、饮食、发病部位等因素与发病几率的关系。结果显示:犬乳腺肿瘤常见于杂交犬、京巴犬及西施犬,多发于母犬,发病平均年龄为9岁,且以未进行绝育手术、平时饲喂高能量食物的犬发生乳腺肿瘤几率较高,发病部位从胸部到腹部呈递增趋势,且在第3、4、5对乳头发病率较高。2.乳腺肿瘤病理组织学研究:取手术摘除的犬乳腺肿瘤置于福尔马林溶液中,运用病理组织学方法,制作病理切片于光学显微镜下观察,结果显示:犬恶性肿瘤发病率较高,20例患犬中14例为恶性肿瘤,占总乳腺肿瘤的70%,主要有癌肉瘤、浸润性导管癌、乳腺小叶原位癌、单纯癌和腺癌。良性肿瘤病例有6例,占总乳腺肿瘤的30%,主要为有导管内乳头状瘤和纤维性病。3.犬乳腺肿瘤ER、PR和c-erbB-2检测:取置于多聚甲醛溶液中的组织样本,采用免疫组织化学方法检测雌激素受体、孕激素受体及表皮生长因子受体的表达情况,结果显示:ER、PR在良性肿瘤中的阳性表达率均高于恶性肿瘤,c-erbB-2阳性表达检测中,恶性肿瘤的阳性表达率要高于良性肿瘤。本实验研究证明犬类乳腺肿瘤也可能存在类似于人类乳腺肿瘤中“三阴乳腺癌”的发生机理,根据犬乳腺肿瘤及其相关因子检测结果更进一步说明犬对人类乳腺肿瘤病理研究具有重要参考价值。
[Abstract]:Breast tumor is one of the most common tumors in dogs. Its high incidence rate and high mortality rate seriously endanger the health of dogs. Although the prevention and treatment of canine breast tumors has been improved and improved in recent years, the cure rate has not been significantly improved. The main reason is that the diagnosis of the disease is of great significance to the treatment of the disease. As a companion of human beings, the occurrence of mammary gland tumors in dogs has a certain reference value for the study of human breast tumors. The diagnosis of canine mammary neoplasms and the detection and analysis of related predictors were studied. 1. Investigation and analysis of clinical cases of canine breast neoplasms: the history of canine disease was investigated, the clinical symptoms and the process of diagnosis and treatment were observed, and the breeds, sex, age, fertility status, diet of the affected dogs were statistically analyzed. The relationship between the location of the disease and the incidence of the disease. The results showed that breast neoplasms were common in hybrid dogs, Jingba dogs and Xi Shi dogs, and mostly in female dogs. The average age of the disease was 9 years old. The incidence of nipples increased from chest to abdomen, and the incidence of nipples was higher in the 3rd 5th tit. Histopathological study of Breast tumor: the canine breast tumor was removed and placed in formalin solution. The pathological sections were made by using histopathological method and observed under optical microscope. The results showed that the incidence of malignant tumors in dogs was higher. 14 of the 20 cases were malignant tumors, accounting for 70% of the total breast tumors, including carcinosarcoma, invasive ductal carcinoma, breast lobule carcinoma in situ, simple carcinoma and adenocarcinoma. There were 6 cases of benign tumors, accounting for 30% of the total breast tumors, mainly intraductal papilloma and fibrous venereal diseases. Detection of ERP PR and c-erbB-2 in canine breast neoplasms: the expressions of estrogen receptor, progesterone receptor and epidermal growth factor receptor were detected by immunohistochemical method. The results showed that the positive expression rate of PR in benign tumors was higher than that of c-erbB-2 in malignant tumors, and the positive expression rate in malignant tumors was higher than that in benign tumors. This study has proved that the mechanism of "three yin breast cancer" in human breast cancer may also exist in canine breast tumors. According to the detection results of canine mammary neoplasms and its related factors, it is further proved that dogs have important reference value for the study of human breast neoplasms.
