番鸭MC1R基因多态性与羽毛相关性分析

【摘要】:为探讨黑素皮质素受体1(melanocortin-1 receptor,简称MC1R)基因对番鸭羽色的影响,采用PCR和直接测序法对番鸭MC1R基因编码区进行多态性分析,比较不同羽色番鸭群体遗传参数和基因型分布卡方检验。结果表明:在番鸭MC1R基因编码区发现G274A、G279A、C485A突变,使第92处氨基酸由谷氨酸(E)变为赖氨酸(K),第162处的氨基酸由苏氨酸(T)变为天冬氨酸(N)。通过各个位点不同羽色番鸭群体基因型、等位基因频率的比较分析,同时结合不同群体间基因型分布卡方检测,发现在第279处的G-A突变中白羽番鸭仅有GG型,而其他3个携带大量黑羽的番鸭群体都以GA、AA型存在;同时全白番鸭与其他3个番鸭群体之间卡方值最大,推测此位点对番鸭的羽色影响较大。而从其他2个突变位点的数据分析推测,它们对番鸭的羽色几乎没有影响。研究结果为开展番鸭羽色研究和育种工作奠定了基础。
[Abstract]:In order to study the effect of melanin receptor 1(melanocortin-1 receptor (MC1R) gene on the feather color of muscovy duck, PCR and direct sequencing were used to analyze the polymorphism of MC1R gene coding region in muscovy duck. The genetic parameters and genotype distribution of different feather muscovy duck populations were compared by chi-square test. The results showed that in the MC1R coding region of muscovy duck, the mutation of G274An G279AN C485A was found, which changed the 92nd amino acid from Glutamic acid E) to Lysine Ko, and the 162nd amino acid from threonine to aspartic acid. Based on the comparative analysis of genotype and allelic frequency of different loci in different populations of Muscovy duck, combined with Chi-square analysis of genotype distribution among different populations, it was found that White Muscovy Duck had only GG type in G-A mutation at 279. The other three muscovy duck populations with a large number of black feathers were GAAA-type, and the chi-square value between the whole white muscovy duck and the other three muscovy duck populations was the largest, which suggested that this locus had a great influence on the feather color of muscovy duck. From the data analysis of the other two mutation sites, it was inferred that they had little effect on the feather color of muscovy duck. The results laid a foundation for the research and breeding of Muscovy duck feather color.
