妊娠后期IUGR对蒙古绵羊胎儿肾脏生长发育的影响

【摘要】:本实验研究了蒙古绵羊妊娠后期营养限饲引起胎儿IUGR,对胎儿肾脏生长发育的影响。选择体况中等、2-3胎次,同期发情及受孕的健康蒙古绵羊24只,妊娠期90d时,选出6只母羊开始屠宰实验,其余按体重任意分配到能量水平依次降低的3组,其中每组6只。饲喂在不同的能量饲养水平:CG组(自由采食,0.67MJME·BW-0.75·d-1);RG2组(0.33 MJME·BW-0.75·d-1);RG1组(0.175MJME·BW-0.75·d-1),至妊娠140d时对全部实验组母羊进行屠宰。实验结果表明:(1)妊娠后期随着营养限制程度加大,RG1组(P0.01)和RG2(P0.05)胎儿肾脏重和肾脏生长速率明显降低;RG2、RGl组(P0.05)胎儿肾脏干物质重和脂肪重显著降低;RG1组水分重和蛋白质重显著降低(P0.01);RG2(p0.05)、RG1组(p0.01)灰分重低于CG组。(2)妊娠后期随着营养限制程度加大,单位视野内,RGl组肾小球的数目多于RG2组(P0.05),系膜轻度增生,增加系膜细胞数量,毛细血管袢明显增厚;Masson染色胶原蛋白纤维化程度呈加深趋势,RG1组I胶原蛋白浓度极显著低于CG组(P0.01)。(3)妊娠后期随着营养限制程度加大,RG1、RG2组胎儿肾脏组织中总DNA含量显著低于CG组(P0.01)。(4)妊娠后期随着营养限制程度加大,RG1胎儿肾脏T-AOC低于CG组(P0.05组;RGl组胎儿肾脏中SOD活性显著低于CG组(P0.05);RG1组胎儿肾脏中GSH-Px活性显著低于RG2(P0.05)、CG组(P0.01):RG1组胎儿肾脏中MDA的浓度均显著高于RG2、CG组(P0.01);RG1组抑制羟自由基能力高于CG组(P0.01);RG1(P0.01)、RG2组(P0.05)胎儿肾脏黄嘌呤氧化酶活力低于CG组;RG1组胎儿肾脏过氧化物酶活力低于CG组(P0.01)(5)妊娠后期随着营养限制程度加大,RG1胎儿肾脏Scr和BUN低于CG、RG2组(P0.01);RG1组胎儿肾脏中β2-MG显著高于CG、RG2组组(P0.05);RG1组胎儿肾脏中Cys C显著高于于CG组(P0.01)。
[Abstract]:The effects of feeding restriction on fetal kidney growth in Mongolian sheep were studied. At 90 days gestation, 6 ewes were selected to begin slaughtering experiment, and the others were divided into 3 groups according to their weight and energy level, among which 6 were in each group. All the ewe of experimental group were slaughtered at different energy levels (0.67 MJME BW-0.75 d-1 + RG2 0.33 MJME BW-0.75 d-1) and 0.175 MJME BW-0.75 d-1 at day 140 of gestation. The results showed that the fetal kidney weight and kidney growth rate decreased significantly in the late pregnancy with the increase of nutritional restriction (P0.01) and RG2P0.05) the weight of dry matter and fat of fetal kidney decreased significantly in RG1 group and protein in RG1 group. The ash content in RG1 group was significantly lower than that in CG group (P 0.01). The number of glomeruli in the RGl group was more than that in the RG2 group (P 0.05), and the Mesangial cells were slightly proliferated and the number of Mesangial cells was increased. The degree of collagen fibrosis in RG1 group was significantly lower than that in CG group (P 0.01. 01. 3) the total DNA content in renal tissue of RG1RG2 group increased with the increase of nutritional restriction in the latter trimester of pregnancy. Lower than CG group (P0.01P0.01P0.01P0.01P0.01, P < 0.05) the SOD activity of RG1 fetal kidney in late pregnancy was lower than that of CG group (P 0.05). The SOD activity of RG1 group was significantly lower than that of CG group (P0.05 + RG1 group was lower than that of CG group P0.01 + RG1 group.) the fetal kidney SOD activity in RG1 group was significantly lower than that in CG group (P0.01 + RG1 group). The concentrations of MDA in the viscera were significantly higher than those in RG2CG-P0.01P0.01RG1 group. The ability of inhibiting hydroxyl free radicals in RG1 group was higher than that in CG group. The activity of xanthine oxidase in fetal kidney was lower than that in CG group. The activity of renal peroxidase in RG1 group was lower than that in CG group. The levels of Scr and BUN in RG1 fetal kidney were significantly higher than those in CG group (P 0.01). The level of 尾 2-MG in RG1 group was significantly higher than that in CG group (P 0.05) and that in RG 1 group was significantly higher than that in CG group (P 0.01).
