3号中草药添加剂对仔猪免疫功能影响的研究

【摘要】:中草药具有毒副作用小,绿色无残留的特点,已成为国内饲料添加剂研究领域的热点,尤其是中草药添加剂提高动物机体抗病能力的研究,越来越受到人们的关注。为探讨3号中草药添加剂对仔猪部分主要免疫指标的影响,为提高仔猪抗病力和养猪业的绿色健康发展提供科学依据。试验随机选取15日龄左右经检测Mhp抗体为阴性的仔猪60头,随机分为对照组、中草药组、疫苗组、中草药+疫苗组,连续饲喂3号中草药10d,分别进行了3号中草药添加剂对仔猪支原体肺炎灭活疫苗免疫效果,部分血常规和血液生化指标,血清中IL-2、IL-4、IFN-γ的含量,仔猪T淋巴细胞亚群和增殖影响的试验。结果如下:1、3号中草药添加剂对仔猪支原体肺炎灭活疫苗免疫效果的影响为探讨3号中草药添加剂对仔猪灭活疫苗免疫效果的影响,试验对疫苗组和中草药+疫苗组的仔猪15日龄首免安百克灭活疫苗,并与30日龄二免,分别在实验前、30日龄和50日龄采集血清,采用ELISA方法检测猪支原体肺炎抗体水平。结果:30日龄和50日龄时,疫苗组和中草药+疫苗组的S/P值均高于空白对照组和中草药(P0.01);中草药+疫苗组的S/P值显著高于疫苗组(P0.05)。显示:3号中草药添加剂能够提高仔猪支原体肺炎灭活疫苗免疫效果。2、3号中草药添加剂对仔猪部分血常规和血液生化指标的影响为探讨3号中草药添加剂对仔猪部分血常规和血清生化指标的影响,试验分别于实验前、30日龄和50日龄采血,采用全自动动物血细胞分析仪和血液生化分析仪进行检测,结果:30日龄时,与对照组相比:中草药组、疫苗组、中草药+疫苗组的WBC分别提高了4.75%、8.72%、18.62%(P0.05);LYM分别提高了9.83%、15.08%、25.57%(P0.05);Gran分别提高了15.12%、21.00%、36.12%;RBC分别提高了8.36%、2.85%、9.12%;TP分别提高了15.81%(P0.05)、6.67%、16.85%(P0.05);ALB分别提高了6.37%、-0.74%、10.61%;GLO分别提高了38.71%(P0.05)、24.61%、32.00%(P0.05)。50日龄时,与对照组相比:中草药组、疫苗组、中草药+疫苗组的WBC分别提高了8.39%、34.54%、36.45%;LYM分别提高了28.80%、12.58%、31.78%;Gran分别提高了12.23%、0.53%、29.25%;RBC分别提高了11.45%、7.21%、27.23%(P0.05);TP分别提高了14.87%(P0.05)、12.30%(P0.05)、23.45%(P0.05);ALB分别提高了11.08%、6.59%、14.07%(P0.05);GLO分别提高了22.81%、24.27%、43.10%(P0.05)。表明:3号中草药能够提高仔猪外周血中白细胞、淋巴细胞、红细胞的数量和清中总蛋白、白蛋白和球蛋白的含量。3、3号中草药添加剂对仔猪血清中IL-2、IL-4、IFN-γ含量的影响为探讨3号中草药添加剂对仔猪血清中IL-2、IL-4、IFN-γ含量的影响,试验采用ELISA试剂盒进行了检测,结果:30日龄时,与空白对照组相比:中草药组、疫苗组、中草药+疫苗组IL-2的浓度分别提高了0.92%、4.14%、5.32%(P0.05);IL-4的浓度分别提高了3.44%、14.22%(P0.05)、12.18%(P0.05);IFN-γ的浓度分别提高了4.11%、6.55%、21.12%(P0.05)。50日龄时,与空白对照组相比:中草药组、疫苗组、中草药+疫苗组IL-2的浓度分别提高了4.10%(P0.05)、1.05%、12.75%(P0.05);IL-4的浓度分别提高了7.63%、7.42%、8.54%;IFN-γ的浓度分别提高了4.15%、4.06%、12.44%(P0.05)。说明:3号中草药添加剂提高了仔猪血清中IL-2、IL-4、IFN-γ的浓度,尤其是显著提高了中草药+疫苗组的IL-2、IL-4、IFN-γ含量。4、3号中草药添加剂对仔猪T淋巴细胞亚群及T淋巴细胞体外增殖的影响为探讨3号中草药添加剂对仔猪T淋巴细胞亚群及T淋巴细胞体外增殖的影响,试验采用流式细胞术检测仔猪外周血T淋巴细胞亚群CD4~+、CD8~+的百分比变化;采用CCK-8法进行了T淋巴细胞体外增殖试验。结果:30日龄时,与对照组相比:中草药组、疫苗组、中草药+疫苗组的CD4~+T细胞百分比分别提高了8.96%、16.91%、26.24%;CD8~+T细胞百分比分别提高了3.33%、4.62%、7.02%(P0.05);CD4~+/CD8~+的值分别增长6.88%、11.85%、19.41%(P0.05);T淋巴细胞刺激指数(SI)分别增长了5.47%,3.48%和10.02%(P0.05)。50日龄时,与对照组相比:中草药组、疫苗组、中草药+疫苗组的CD4~+T细胞的百分比分别提高了13.43%、10.47%、33.30%(P0.05);CD8~+T细胞百分比分别提高了2.42%、5.00%、-2.21%;CD4~+/CD8~+的值分别提高了14.87%、6.49%、32.68%(P0.05);T淋巴细胞刺激指数(SI)分别增长了6.90%,2.24%和9.80%(P0.05)。表明:3号中草药添加剂能够提高仔猪外周血中CD4~+T细胞百分比、CD4~+/CD8~+的比例和T淋巴细胞体外增殖能力,尤其是能显著提高中草药+疫苗组的CD4~+T细胞、CD8~+T细胞的百分比以及CD4~+/CD8~+的比例和T淋巴细胞刺激指数(SI)。因此,3号中草药能够提高仔猪支原体灭活疫苗的免疫效果,血液中白细胞、红细胞和淋巴细胞的数量,血清中总蛋白、白蛋白、球蛋白、IL-2、IL-4、IFN-γ的含量,CD4~+T细胞的百分比和CD4~+/CD8~+的值和T淋巴细胞的体外增殖能力,从而提高了仔猪的免疫功能和抗病能力。
[Abstract]:Chinese herbal medicine has the characteristics of small toxic and side effects and no residue of green, which has become a hot spot in the research field of domestic feed additives, especially the study on improving the disease resistance of animal body by Chinese herbal medicine additive, which has attracted more and more attention. A scientific basis was provided for the green and healthy development of the disease power and the pig industry. The test randomly selected 60 piglets with negative Mhp antibody for 15 days of age and randomly divided into the control group, the Chinese herbal medicine group, the vaccine group, the Chinese herbal medicine + vaccine group, and the continuous feeding of the Chinese herbal medicine 10d, which was used to inactivate the piglet mycoplasma pneumonia inactivated vaccine. The immune effect, some blood routine and blood biochemical indexes, the content of IL-2, IL-4, IFN- gamma in the serum, the effects of the T lymphocyte subsets and proliferation of piglets. The results are as follows: the effect of the Chinese herbal additive on the immune effect of the piglet mycoplasma pneumonia inactivated vaccine is the shadow of the effect of the 3 Chinese herbal additive on the inactivated vaccine of piglet inactivated vaccine. It indicated that the herb additive in 3 was able to increase the peripheral blood of piglets. The percentage of CD4~+T cells, the proportion of CD4~+/CD8~+ and the proliferation ability of T lymphocytes in vitro, especially the CD4~+T cells, the percentage of CD8~+T cells and the proportion of CD4~+/CD8~+ and the T lymphocyte stimulation index (SI) in the Chinese herbal + vaccine group. Therefore, 3 Chinese herbal medicine can improve the immune effect of the piglet inactivated vaccine. The number of leukocytes, red cells and lymphocytes in the liquid, the content of total protein, albumin, globulin, IL-2, IL-4, IFN- gamma in serum, the percentage of CD4~+T cells and the value of CD4~+/CD8~+ and the ability of T lymphocyte proliferation in vitro, thus improve the immune function and disease resistance of piglets.
